Objective: This experiment was conducted in the general condition of diseased rats with ulcerative colitis(UC),disease activity index(DAI)score,gross observation of colon tissue,pathological changes,serum GSH,GSH-Px in intestinal supernatant,and serum IFN-? expression.To investigate the mechanism of Huazhujieduyuyang decoction in the treatment of UC,and provide experimental basis for clinical application.Method: 60 clean-tested male Wistar rats(200g-220g)were fed adaptively for 7 days and randomly divided into 6 groups: normal control group(NG),model group(MG),and mesalazine group(MSG).,Huazhuojiedu Yuyangjian high dose group(ZHG),middle dose group(ZMG),low dose group(ZLG),10 in each group.Except for normal feeding and no treatment in NG group,all other groups were modeled by TNBS/alcohol compounding method.On the 4th day after successful model establishment,MG group was given 4 ml/kg physiological saline once a day,MSG group daily.The mesalazine suspension was intragastrically administered once,and the Chinese medicine group was given gavage with a corresponding concentration of Huazhuo Jiedu Yuyang Decoction once daily.After 14 days of continuous intervention,the general condition of rats was observed(reaction sensitivity,dietary intake,Hair gloss,fecal traits,body weight changes,and DAI scores.After the end of treatment,the rats were sacrificed by decapitation.The blood was taken from the femoral artery.The colon tissue was dissected and cut into two parts along the longitudinal axis.Some tissues were paraffin-embedded,stained with HE,and the pathological conditions of the mucosa were observed under electron microscope.A portion of the colon tissue was used to separate the supernatant.Serum IFN-? levels were measured by enzyme-linked immunosorbent assay.Serum GSH and GSH-Pxlevels in colon tissue supernatants were detected by biochemical methods.Results:1.General situationThroughout the experiment,rats in the NG group had the best overall condition,with well-groomed and shiny coats,responsiveness,rapid activity,and ecstatic feeding.The stools were dark granular,and the perianal area was clean and weight increased.The rest of the rats in each group showed varying degrees of loose stools and mucous pus and blood after the model was established.The perianal contamination was filthy,the Xiza piled up lying,the coat was messy and dull,and did not like feeding,and the spirit was poor.After drug intervention,the rats in the ZHG,ZMG,ZLG,and MSG groups all showed improvement.The ZHG group had the most obvious improvement,but the rats in the MG group were deteriorating,with apathetic mood,loss of appetite,perianal turbidity,and mucopurulent blood.Increased weight loss.2.DAI scoreZHG group,ZMG group,ZLG group and MSG group were all lower than MG group,the difference was statistically significant(P<0.05);ZHG group was lower than ZMG group,ZLG group,and MSG group,the difference was statistically significant(P<0.05).ZMG group and MSG group were lower than ZLG group.The difference was statistically significant(P<0.05).There was no significant difference between MSG group and ZMG group(P<0.05).3.Colon histopathological observationIn the NG group,the colonic epithelial surface was intact,with a large number of goblet cells.The crypts were straight and long without distortion and branching.The mucosal layer was not thickened.There was no edema and vasodilatation in the submucosa.Intrinsic membrane was occasionally seen inflammation.cell.In the ZHG group,the colon surface was intact and uninterrupted,and a small amount of inflammatory cells infiltrated in theintrinsic membrane.The crypts were longer but branched.There was no significant difference between the ZMG group and the MSG group.There were partial loss of tissue epithelium,more goblet cells,and focal lymphoid and plasma cell infiltration in the lamina propria.In the ZLG group,the mucosal degeneration was obvious,the glands were poorly arranged,the goblet cells were significantly decreased,and the muscular layer was thickened.The inflammatory cells in the lamina propria were flaky.The rats in the MG group had mucosal erosions and ulcers,and the surface covered with white moss.The necrotic tissue and numerous inflammatory exudates were seen under the microscope.4.Indicator detectionGSH expression: The serum GSH levels in ZHG,ZMG,ZLG,MSG,and MG groups were lower than those in NG group(P<0.05);ZMG,ZLG,MSG,and MG groups were lower than ZHG group,and the difference was statistically significant.(P<0.05);ZLG and MG groups were lower than ZMG group and MSG group,the difference was statistically significant(P<0.05);there was no significant difference between ZMG and MSG group(P>0.05);MG was lower In the ZLG group,the difference was statistically significant(P<0.05).GSH-Px expression: NG group was higher than ZHG,ZMG,ZLG,MSG and MG groups,the difference was statistically significant(P<0.05);ZHG group was higher than ZMG,ZLG,MSG and MG group,the difference was statistically significant(P<0.05);ZMG and MSG groups were significantly higher than ZLG and MG groups(P<0.05);ZMG and MSG groups had no significant difference(P>0.05);ZLG group was higher than MG In the group,the difference was statistically significant(P<0.05).Expression of IFN-?: The expression of serum IFN-? in ZHG,ZMG,ZLG,MSG and MG groups was higher than that in NG group(P<0.05);The five groups of ZHG,ZMG,ZLG,MSG and MG Compared with ZHG group,the content was the lowest,the difference was statistically significant(P<0.05);there was no significant difference in serum IFN-? content between ZMG andMSG group(P>0.05);the content of ZMG and MSG was lower than ZLG and MG.The difference was statistically significant(P<0.05);the ZLG group was lower than MG group,and the difference was statistically significant(P<0.05).Conclusion: Huazhuo Jieduyuyangyang decoction can effectively improve the general condition of rats with ulcerative colitis and reduce the DAI score.The high-dose Chinese medicine group has the best curative effect,and the mechanism may be reduced by reducing the content of proinflammatory cytokine IFN-?.Inflammatory response,while increasing the content of GSH and GSH-Px with antioxidant effects,scavenging oxygen free radicals,protecting tissues from oxidative stress damage,and maintaining the body's normal immune regulation.
