Objective:Salsola collina,a perennial herb of the Chenopodiaceae,is widely distributed in various regions of China,and has been used as folk treatment on hypertension.According to“Anhui herbal”records,Salsola collina could cure habitual constipation,but there are few studies on this aspect.In the present stugy,the extraction process,gastrointestinal motility and antioxidant activities of Salsola collina were studied,and effervescent tablets were prepared.Methods:Salsola collina was extracted by the ethanol refluxing method and the effects of single factor such as ratio of liquid to material,ethanol concentration,reflux time on the yield of extract were studied.The extraction process was optimized by response surface method to obtain the ethanol extract.The different fraction extracts were obtained by sequential extraction with petroleum ether,ethyl acetate and n-butanol.The effects of different fraction extracts on gastric emptying,small intestinal propulsion and contractile activity of isolated gastric smooth muscle strips in normal rats were detected,and the main active fraction was determined.The effects of ethyl acetate extract on gastric emptying,intestinal propulsion and isolated gastric smooth muscle strips in rats with gastrointestinal motility disorders induced by atropine and adrenalin were observed.ELISA was used to determine the effect of ethyl acetate extract on motilin,ghrelin and gastrin secretion in normal rats,thus the preliminary mechanism of promoting gastrointestinal motility was explored.The antioxidant activities of different extracts were tested by hydroxyl and oxygen radical scavenging and reducing power.The ethyl acetate extract was used as the main component for effervescent tablets,and were made by acid-base separated aqueous granulation.The formula was optimized by orthogonal design based on disintegration time,pH,foaming capacity and sensory assessment.Results:The optimal extraction conditions of Salsola collina were ratio of liquid to material 20mL/g,ethanol concentration 70%,reflux time 2.25h,and the yield of the extract was 13.56%.The effect of ethyl acetate extract on gastric emptying and intestinal propulsion in normal rats was most significant,and gastric emptying and small intestinal propulsion rate were 67.67±3.85%and 74.15±3.12%?compared with the saline group,P<0.01;compared with the positive drug,P>0.05?.The ethyl acetate extract could improve the gastrointestinal motility retardation induced by cisplatin.The gastric emptying rate increased from 40.07±2.55%to 50.86±3.15%,and the small intestinal propulsion rate increased from 50.58±3.13%to 56.48±3.14%?compared with the saline group,P<0.01?.The ethyl acetate extract could resist gastric emptying and intestinal propulsion in rats with gastrointestinal motility disorder induced by atropine?P<0.01?,but could not resist gastric emptying and intestinal propulsion in rats induced by epinephrine?P>0.05?.The ethyl acetate extract could increase the level of ghrelin and gastrin in plasma of rats?P<0.01 and P<0.05?,but not increase motilin level?P>0.05?.The contractile effects of ethyl acetate extract on isolated gastric smooth muscle strips of rats were most significant?compared with the saline group,P<0.01?.The ethyl acetate extract could significantly improve gastric smooth muscle relaxation induced by atropine and epinephrine?P<0.01?.Furthermore,the extracts of Salsola collina had antioxidant effects.The hydroxyl radical scavenging,superoxide anion radical scavenging and reducing power of ethyl acetate extracts were 77.94±3.26%,65.94±3.95%and 0.83±0.05 at concentration of0.5mg/mL,respectively.The removal rate is secondary to Vc while the reducing power is equivalent to Vc.The optimum formulation of effervescent tablets as follows:12.0%salsola collina extract,45%effervescing agent with tartaric acid-NaHCO3 ratio of 1:2.5,and 2.5%aspartame.The content of total flavonoids was 10.16±0.25mg/g.Conclusions:Salsola collina extracts had the effects of promoting gastrointestinal motility and antioxidant,and the ethyl acetate extract is the main active site.The mechanism of promoting gastrointestinal motility is not single which might affect M-cholinergic receptor and adrenergic receptor pathway and increase plasma ghrelin and gastrin.The formula of effervescent tablets is simple and feasible,and the obtained tablets conform to the requirement of effervescent tablets.
