Objective After observing the radiation injury of spinal cord nerve cell(SCN),we explored the possible mechanism of Lycium barbarum polysaccharides(LBP)by inducing autophagy,and provided a new direction for protecting the organism after radiation injury.Methods After cultured in vitro,SCN of spinal cord neurons was irradiated with different doses(2,6,10Gy)X ray.Detection of cell survival rate by MTT method and determination of optimal radiation dose.Different concentrations(15,25,40mg/L)of LBP were used to interfere the radiation damage of SCN cells.24 h and MTT were used to detect cell survival rate,and the best drug intervention concentration was used to detect cell viability and determine the best radiation dose.A radiation injury model was determined.The experiment was divided into three groups,normal control group,radiation injury group and radiation +LBP group.The number of autophagy lysosomes was observed by electron microscope,and the expression of LC3 II / I and Beclin-1 of autophagy related genes were detected by western blot and immunohistochemistry.Results Radiation damage can reduce the survival rate of spinal cord nerve cells,MTT method was used to detect different X-ray dose of the survival rate of SCN was 85.00+ 3.91,70.00+ 1.63,49.25 + 7.59,visible X ray radiation,the survival rate of SCN cells in a certain range with the increase of the dose decreased,the difference was statistically significant(P<0.05).MTT results showed that different concentrations(15,25,40mg/L LBP)on the radiation damage of the cell survival rate of SCN is 57.25 + 3.09,60.25 + 5.56,70.25 + 2.87,49.25 + 7.59 and radiation group,cell survival rate has improved signifiantly,with statistically significant difference(P<0.05).The concentration of 40mg/L LBP on prognosis afterradiation injury of SCN stem cells,electron microscope can observe the autophagy lysosome increased,while +LBP group was significantly higher than the radiation injury group more,Western blot and immunohistochemistry results showed that: normal group,radiation group,+LBP radiation group,the expression of gene LC3 II / I and Beclin-1 there were increased,with statistical significance difference(P<0.05).Conclusion Lycium Barbarum Polysaccharide has protective effect on radiation injury of cultured spinal cord neurons in vitro.
