In recent years,with the improvement of the social,economic,and environmental benefits of succulents,the market is in short supply.Therefore,based on the variety characteristics of succulent plants,it is necessary to explore several kinds of efficient techniques for rapid propagation of more fleshy meat,and to form a set of succulent plant asexual propagation technology system so as to bring greater economic and social benefits is imperative.In this study,firstly from the improvement of the culture medium of succulent plants,the research on the cutting propagation and tissue culture of Sedum,Echeveria Glauca gigantea,Graptopetalum amethystinum,Cotyledon tomentosa,Haworthia cooperi var.picturata,Sun Moon Lake and Haworthia cymbiformis fvariegata were studied.By studying the effects of different cutting substrates,different hormones,different treatment time of hormones,and different cutting sites on their growth,we obtained the best and fastest breeding treatments for these succulents.After that,the tissue culture of the succulent plants with unfavorable cutting propagation conditions was studied.Different hormones,different media,different concentrations of the same hormone,different light qualities,and different parts of the plant were studied.The aim is to obtain the most ideal medium,increase its survival rate,thereby increase its propagation coefficient,and provide certain technical and theoretical references for the rapid propagation of other succulent plants.The main results of this experiment are as follows: 1.Cutting propagation studies:a.Different cutting substrates have influence on the propagation of succulent plants of Sedumaceae,and found that the survival rate of matrix M4 is the highest and that of more meat is best.b.Studies on the same substrate by different succulents found that the survival rate of different plant species is also different.Survival rate: Sedum & more meat> Liliaceae,in which the survival rate of various plants,Echeveria Glauca gigantea > Sedum morganianum > Cotyledon tomentosa,peach eggs> Haworthia cooperi var.picturata,Sun Moon Lake.The survival rate of leaf intercropping of the succulents of Liliaceae is 0.The succulents of Liliaceae should not use leaf propagation.c.The same hormone IBA treatment at different time,the time is too long to inhibit the rooting of the meat.The IBA treatment had the best overall effect when the bead curtain time was 40 s.For the Cotyledon tomentosa and the peach eggs,the best effect was achieved when the overall effect was 80 s.d.Treating several succulents with different hormones,6BA-NAA has the best combination effect;6-BA has the worst effect.Hormones inhibit the growth of Cotyledon tomentosa.The best rapid propagation method of succulent plant cuttings was as follows: leaf and stem + matrix M4 + suitable hormone treatment time + 6BA-NAA root,improving the survival rate of multi-meat cuttings.2.Tissue culture studies: Primary culture,a.Treatment of several succulents with different hormones,Formula 4 is the best;b.Treatment of different culture media based on Formula 4 showed that the 3/4 MS medium had the highest callus induction rate and the callus induction effect was ideal.c.Adding different concentrations of hormone KT on the basis of Formula 4,the differences from the largest to the smallest were peach eggs> Cotyledon tomentosa > Chrysanthemum(Haworthia cymbiformis fvariegata),Sun Moon Lake,and Momieshou.It has no significant influence on the callus formation of Chrysanthemum(Haworthia cymbiformis fvariegata),Sun Moon Lake,and Momieshou.The formula D has the best induction effect.d.Different light quality treatments had the highest white light injury induction rate and the best callus induction effect,followed by red,blue,blue,and red.e.The induction rate of the callus was significantly different in different parts of the plant,and the stem induction effect was the best.Leaf and stem> stem> leaf> root.f.The effects of different cutting methods on the callus of plants were significantly different.Longitudinal and oblique> crossed.The best way to increase the rate of callus production is: 3/4MS medium + formula 4 + white light + flower stems or leaves chamfering.Subculture proliferation and differentiation culture: a.Different hormones treatment extinction Momieshou,Sun Moon Lake,Chrysanthemum(Haworthia cymbiformis fvariegata),the formula 4 is the best effect of callus proliferation and differentiation.b.Treatment of different culture media based on Formula 4 showed that the 3/4 MS medium had the highest callus induction rate and the best callus proliferation and differentiation effects.c.Different concentrations of hormone KT,formula D has the best influence on the proliferation and differentiation of Cotyledon tomentosa;Formula C had the best effect on the proliferation and differentiation of peach eggs,Sun Moon Lake,Chrysanthemum(Haworthia cymbiformis fvariegata),and Momieshou.d.Different light quality treatments yield white light> red blue> blue light> red light.Different light quality treatments had significant differences in the differentiation of peach egg buds.It is inferred that different light qualities have an effect on pigment formation in the body during plant growth.e.Proliferation and differentiation of callus in different parts of plants.The difference in proliferation coefficient: stem> leaf base> leaf> leaf tip.The best way to increase its proliferation and differentiation is: 3/4MS medium + formula 4 + white light + flower stem.Different rooting media,research found that A3 rooting medium has the best rooting effect.
