| the research objective: to observe the rats in motility estrus restrain condition ovaries mitochondrial DNA copy number changes and NADH expression level changes.Preliminary discussion from molecular level motor caused by the onset of estrus inhibition mechanism and pathological results.In order to prevention of athletic menstrual disorder and treatment to provide new clues.Methods: health and estrus normal SD rats(3 months of age,female)30 were randomly divided into 2 groups: blank control group(group C,n = 10),exercise group(group E,n = 20).Control group without any intervention,natural feeding until the end of the experiment.Exercise group rats,large load swimming training on a daily basis,train six days a week,one day off.All rats,vaginal smears on a daily basis in order to observe the changes in estrus.When exercise group rats for three consecutive estrus a substantial reduction in vaginal epithelial keratinocytes and even disappear when determine the experiment building success.Building after the success of the general conditions and weight change,observed in the rat sterile extract the ovary,with tested according to the method of Real-time PCR amplification mitochondrial DNA copy number,with a known concentration of plasmid DNA as a standard for standard curve to quantify mitochondrial DNA copy number,analysis of the control and exercise group copy number variations,and to analyze its correlation with the condition of mutation.Using Western blot(Western blot)method to detect the mitochondrial respiratory chain complexes speed limit(NADH)protein expression level.Research results: 1.With the experimental process,exercise group rats oestrous cycle derangements in succession,the activity decreases,unresponsive,listlessness,easily frightened,feeding water reducing,hair sparse lacklustre,slow weight gain.Normal control group rats oestrous cycle,well developed,the active,water ingestion is normal,hair neat luster,weight stable growth.2,the experimental results show that the individual sports group than control group in ovarian mtDNA relative copy number were significantly lower(p < 0.01).Control group average mtDNA relative copy number was 82.74 + /-3.85,exercise group average mtDNA relative copy number of only 38.54 + /-1.42,average two groups showed significant difference(p < 0.01).3,will the statistical sample NADH(51 kd)and its corresponding GAPDH internal protein(36 kd)two grey value compared to income ratio for statistical analysis.Control group NADH average gray level ratio was 0.89 + /-0.17,and the value was 0.36 + /-0.09 in the exercise group,extremely significant difference(p < 0.01).Research conclusions:1,long-term heavy load movement against the general situation and the development of viscera of rats,to make the body in the process of anaerobic stimulation for a long time,cause ovarian pathological changes in the mitochondrial structure and function,may result in rats oestrous cycle.2,long-term load movement affects the self-replicating features of mtDNA,exercise group rats ovaries mtDNA copy number decreased significantly,suggest long-term high intensity training inhibition of the mitochondrial DNA replication.3,long-term load exercise prevent the transcription of mtDNA function,reduce the enzyme activity of mitochondria,decreased respiratory chain enzyme activity.The body low enabled,anaerobic power way for a long time,relatively weakened the mitochondrial oxidative phosphorylation function.This experiment from the aspects of mitochondrial estrus inhibition caused by high intensity exercise for a long time to provide the corresponding experimental data support,for the prevention and treatment of motility amenorrhoea provides new clues to further explore. |
