The camellia seed oil is one of the main varieties of edible oil in China.In recent years,the cold pressing method was advocated to got to pressed camellia seed oil,it usually can extracted out 86%-92%tea oil from raw materials.The cold pressed camellia cake also contains not less 8%of tea oil and 9%-14%tea saponin after cold pressing.Nowdays,the 6#solvent is the mainly extraction solvent.But it's flash point is-30?,it's very dangerous under normal temperature.The flash point is 35? about N-butyl Alcohol,so it's more security than 6#solvent.The N-butyl Alcohol also can dissolution of vitamin,squalene,lecithin and other substances about tea cake,but the 6#solvent van't dissolution this.Camellia oil is rich in polyunsaturated fatty acid,it can regulate blood cholesterol and inhibit issue peroxidation.Tea saponin is a kind of natural non-ionic surface active agent.Tea saponin has good emulsifying,dispersing,wetting,foaming,foam stability and detergency ability.It also has the effects of anti-permeability,anti-inflammation,analgesia and the dilation of blood vessels.It was widely used in daily chemical industry,medicine,food,building materials,metal material processing and other fields.So,it's very important to make full utilized of cold pressed camellia cake.For the camellia seed oil enterprises,it's a very important method for fast detected about the moisture and oil content of camellia seed.The traditional method need a long time and organic reagents.Near infrared reflectance spectroscopy(NIRS)is a physical test technology and widely application on food,oil,medical,feed,crop.NIRS have some advantage,just like no damage,no pollution and fast.In this paper,the problem exists in camellia seed oil processing.Moisture and oil content of camellia seed was modeled by near infrared reflectance spectroscopy(NIRS).investigated the alliance extraction effected of camellia oil and tea saponin by butanol,analyzed the composition of camellia oil and purified the tea saponin by macroporous adsorption resin.The main research conclusions are as follows:(1)It's study on the optimal process condition of alliance extraction camellia oil and tea saponin from cold pressed camellia cake by N-butanol.The cold pressing camellia cake is used as raw material and alliance extraction camellia oil and tea saponin by N-butanol.Through singer factors analysis,a suitable condition about digesting time,numbers,temperature and solid-liquid ratio was obtained.Then the Box—Behnken experimental design and response surface methodology were taken on regression analysis.Through Gas Chromatograph-Mass Spectrometer analysis,the fatty acid composition was measured and compared about the extracted tea oil and cold pressed tea oil.By the experiment,under the temperature 80?,the suitable condition was digesting 2 hours,3 times and solid-liquid ratio 1:1.2,the tea oil extraction yield reached(92.88±1.41)%,the saponin extraction yield reached(43.2±0.94)%.The Gas Chromatograph-Mass Spectrometer analysis statement that main compound about tea oil by digested and pressed was similar.The tea oil that extracted by n-butyl alcohol was great hold the quality of original.(2)It's study on the purification condition of macroporous rasin about crude tea saponin which was alliance extraction by N-butanol.Through Static and dynamic adsorption.the optimum samples adsorption conditions were studied by single factors and orthogonal experiment.So the optimum process parameters were determined.The optimum desorption parameters were found to be:pH value is 7,sample loading velocity 2 BV/h,volume is 50 mL and concentration is 5 mg/mL;the optimal elution conditions were achieved by using 1.8 BV,80%ethanol as elution solvent at a flow rate of 2 BV/h.Tea saponin product purity can reach to 87.73%.(3)moisture and oil content of camellia seed was modeled by near infrared reflectance spectroscopy(NIRS).A predicition model of moisture and oil content about camellia seed was established based on near infrared reflectance spectroscope(NIRS)and stoichiometry.The spectroradiometer was used to collecting spectra in the wavelength range from 950 to 1 650 nm.The best pretreatment method was selected by scan full spectra area,UNSCRAMBLER9.7 and PLS.The best pretreatment method is Original spectrum + Smoothing + S.Golay + SNV for moisture content about camellia seed.the best pretreatment method is Original spectrum + Smoothing+ MSC for oil content about camellia seed.Those two model is feasible,because the correlation coefficent about moisture content reached 0.916 0,RMSEC is 0.350 1,R2 is 0.839 1,the correlation coefficent about oil content reached 0.973 0,RMSEC is 1.814 0,R2 is 0.946 8,The result of T-test is passable while validation method is cross validation.In this paper,a new and feasible method is built to measure moisture and oil content about camellia seed by near infrared reflectance spectroscope(NIRS)and stoichiometry.
