Objectives:Methods and techniques of extraction,purification and biological activities of efficacy components from the Purple Acanthopanax senticosus were studied in the dissertation.The nutritional composition of the tender stem and leaf of the Purple Acanthopanax senticosus and its micropowder were analyzed,and the method and process of extraction and purification of the anthocyanin and total flavonoids were researched.Edibility safety of the micropowder of the Purple Acanthopanax senticosus and the antioxidant activity of purified the anthocyanins and total flavonoids were evaluated.The study aims to provide the reference and theoretical basis on utilization of the Purple Acanthopanax senticosus.Methods:(1)The content of general nutritional components,minerals,amino acids were determinated by GB standard method.(2)Traditional solution method,microwave-assisted method and ultrasonic-assisted method were used to extract anthocyanins and total flavonoids of the Purple Acanthopanax senticosus.Single factor experiment and orthogonal experiment were adopted to determine the optimum methods of extracting total flavonoids and anthocyanin(3)Experiments of static adsorption,desorption,dynamic adsorption and adsorptionkinetics with AB-8 macroporous resin were conducted to investigate the purification process of anthocyanin and total flavonoids from the Purple Acanthopanax senticosus..(4)Vitamin C was used as control,the oxidation resistance of purified anthocyanins and total flavonoids of the Purple Acanthopanax senticosus were evaluated by using DPPH free radical scavenging method.(5)The acute toxicity test was conducted to evaluated edibility safety of the micropowder of the Purple Acanthopanax senticosus.Results:(1)The moisture contents of the tender stem and leaf and its micropowder of the Purple Acanthopanax senticosus were 87.9%and 6.9%,the contents of ash were 1.2%and 7.9%,crude fiber were 2.21%and 5.63%,total carbohydrate were 2.23%and 7.96%,crude protein were 2.74%and 24.6%;2)The amino acid composition of the Purple Acanthopanax senticosus was analyzed and the proportion of the essential amino acids was 41.06%.2)Minerals and vitamins were rich in the Purple Acanthopanax senticosus,especially for K,Mg,Fe,Ca and Zn.The contents of K,Ca,Mg and Fe were 476mg/100g,91.6mg/100g,45.5mg/100g and 1.68mg/100g,respectively,which is higher than general vegetables.Therefore,the Purple Acanthopanax senticosus is a nutritive vegetable and rich sources of dietary fibers,proteins,minerals.(2)Microwave-assisted extraction was the optimum method of extracting anthocyanins of the Purple Acanthopanax senticosus.The optimum process conditions of extracting anthocyanins of the tender stem and leaf was as follows:extraction time 120s,the microwave power 70W,ratio of material to liquid 1:7,60%ethanol as extractant and extraction times 1.In optimum process conditions,the extraction rate of anthocyanin was 0.024%.The optimum process conditions of extracting anthocyanin of micropowder was as follows:extraction time 100s,the microwave power 210W,ratio of material to liquid 1:60,80%ethanol as extractant and extraction times 1.In optimum process conditions,the extraction rate of anthocyanin was 0.141%.Microwave-assisted extraction was the optimizing method of extracting total flavonoids of the Purple Acanthopanax senticosus.The optimum process conditions of extracting total flavonoids of the tender stem and leaf was as follows:extraction time 80s,the microwave power 70W,ratio of material to liquid 1:5,60%ethanol as extractant and extraction times 3.In optimum process conditions,the extraction rate of anthocyanin was 1.52%.The optimum process conditions of extracting total flavonoids of micropowder was as follows:extraction time 100s,the microwave power 70W,ratio of material to liquid 1:80,50%ethanol as extractant and extraction times 3.In optimum process conditions,the extraction rate of total flavonoids was 20.95%.(3)The AB-8 macroporous adsorption resin had better adsorption property of anthocyanins and total flavonoids of the Purple Acanthopanax senticosus.The adsorption rate of anthocyanins and total flavonoids of the tender stem and leaf were 21.53%and 46.47%.The solution(pH 2.5)had the optimizing adsorption ability for anthocyanins and total flavonoids of the tender stem and leaf.The adsorption rate of anthocyanins and total flavonoids of micropowder were 79.64%and 86.43%.The solution(pH 3.5)had the optimizing adsorption ability for anthocyanins of micropowder,but with pH 2.0 for total flavonoids.(4)Anthocyanins and total flavonoids of the Purple Acanthopanax senticosus had strong scavenging activity to DPPH free radicals._Half scavenging concentration(IC50)of Vc,purified anthocyanins and total flavonoids of micropowder of the Purple Acanthopanax senticosus were 1.49 ?g/mL,0.111 mg/mL and 2.48mg/mL.(5)The result of acute toxicity test showed that the death of rats and mice was not found in the period of 14 days,The LD50 of micropowder of the Purple Acanthopanax senticosus was greater than 1Og/kg.bw.Conclusion:The Purple Acanthopanax senticosus is safe to eat as food and rich in nutrients,especially in protein and dietary fiber.It is a good source of plant protein,and has high content of anthocyanin,flavonoids and high antioxidant activity.The Purple Acanthopanax senticosus had higher developing value for special vegetable and health food,and the market of the promising product will be found.
