Acanthopanax senticosus are belonged to the Araliaceae Plant, their roots, stem, leaves have medical values. The flavonoids, which are one of the main active ingredients, are abundant in Acanthopanax senticosus. Flavonoids are compounds derived from the parent nucleus of flavone and widely exist in nature. Traditionally,they have anti-inflammatory, antivirus and antipyretic effect. In recent studies, it was found that they have antioxidant, scavenging free radicals and anti-tumor effects. The study on this subject has great practical significance.Recently, the main methods applied for extracting flavonoids are decoction by water-ethanol, ultrasonic and microwave treatment, which lead to low purity of target product. The extraction methods of flavonoids were investigated. The methods of water-ethanol reflux extraction and supercritical fluid extraction were used in this experiment. the extraction rate of flavonoids as the indicator come to the best process conditions were:extraction temperature is78℃, the concentration of ethanol is60%and the extraction time is3h in the method of ethanol-water reflux extraction. At last the content of Acanthopanax total flavonoids is only23.36%. the extraction temperature is55℃, extraction pressure is40Mpa and the extraction time is120min in the method of supercritical CO2fluid extraction, as the result the target product from Acanthopanax was determined and the content of total flavonoid is31.13%.Recently, macroporous adsorption resin technology has been used widely in herbal extraction. To improve the content of total flavonoid in the extraction, this study intends to exact and purify Acanthopanax Senticosus Harms extraction by macroporous resin adsorption. Based on comparing adsorption and desorption characteristics of macroporous adsorption resins NKA-9, D301, LSA-21, LX-28, for NKA-9resin, one of the best macroporous adsorption resins is chosen and used to do separation and purification studies. The results show that NKA-9resin can adsorb and desorb total flavonoids well. The best parameters of adsorption and desorption are obtained. Respectively:for adsorption flow rate1ml/min, it has no penetration phenomenon when the sample volume is5BV. For desorption:ethanol-water60%, flow rate2ml/min,6.5BV as the eluent. The yield of the flavonoids is79.9%in this method and the content of the flavonids is58.1%in the product. In conclusion, this method is easily operated and can be suitable for the adsorption and purification of the total flavonoids.
