Chlorogenic acid is a kind of important medicinal active material,not only has the function of antibacterial and antiviral,and increased white blood cells,liver and gallbladder,anti-tumor,antihypertensive,scavenging free radicals and excited central nervous system function.Domestic has been mainly from honeysuckle,eucommia leaf,sunflower seed extraction of chlorogenic acid,but the huge demand for the low prices of raw materials and product contrast forcing people to seek more extensive materials.Chlorogenic acid is contained in tobacco,but few reports about the best extraction method of chlorogenic acid in tobacco.Extraction of chlorogenic acid from tobacco can not only broaden the source of chlorogenic acid,but also can increase the added value of tobacco growers,which has important practical significance to the scientific use of tobacco.In view of this,this paper in determining the optimum extraction process of chlorogenic acid in tobacco,comparing with the test content of different varieties and different parts of tobacco chlorogenic acid,provide technical support for the use of chlorogenic acid in tobacco leaf production,and to provide the scientific basis for the development of tobacco use.The main results are as follows:1.To establish a high performance liquid chromatography(HPLC)method to the determination of chlorogenic acid,sample repeated measurement of variation coefficient is 0.02,the standard addition recovery was 100.46%.Measured chlorogenic acid content was 1.324% in Yun 99.Description of the tobacco leaf by high performance liquid chromatography(HPLC)method for content determination of chlorogenic acid is feasible.2.On the water extraction process of chlorogenic acid in tobacco process,on the basis of single factor and orthogonal test,the optimum process conditions is the temperature of 60 ?,extracting time is 70 min,pH value is 5,and the material ratio of 1:8.The best conditions of chlorogenic acid yield was 1.741%.3.Research on the alcohol extraction process of chlorogenic acid in tobacco process,on the basis of single factor and orthogonal test,the optimum processconditions is 80 ?,extracted two times,extraction time is 90 min,pH value is 5,and the material ratio of 1:12.The best conditions of chlorogenic acid yield was 1.762%.By comparing the two methods,the solvent cost is low and no pollution,which is suitable for the mass production in industry.4.According to the experiment the optimum method were determined for different varieties and different leaf position of chlorogenic acid content in tobacco leaves,the results show that the test material in different parts of the content of chlorogenic acid from big to small order for upper leaves>middle leaves> lower leaves>tobacco stems.In addition,different regions of the same species,Luonan Yun99 of upper leaves,middle leaves,lower leaf of chlorogenic acid are 1.145%?1.004%?0.929%,Nanzheng Yun 99 of upper,middle and lower content of tobacco leaf are1.553%?1.369%?1.143%,compared to Nanzheng cloud 99 chlorogenic acid content is higher in Luonan cloud 99.
