The Extraction Of Geniposide By [Choline][Amino Acid] Ionic Liquid And The Preparation Of Gardenia Blue Pigment

Geniposide not only can be used in medicine,but also can be transformed into gardenia blue pigment by ?-glucosidase.Geniposide is usually extracted using ethanol as solvent in industrial production,whichhas large solvent consumption and environmental problemsIn.Geniposide was extracted by [Choline] [Glutamic acid] Lonic Iiquid([Ch][Glu] ILs)aqueous,then it with the amino acids in the extrac was directly transformed into gardenia blue pigment.The main contents are as follows:(1)Microwave assisted extraction of geniposide by [Ch][Glu] ILsThe extraction conditions of geniposide with aqueous solution of [Ch][Glu] ILs were optimized by response surface methodology.The concentration of [Ch][Glu] ILs was 1.5%,the ratio of liquid to material was 25(mL/g),the microwave power was 320 W,and the time of microwave radiation was 300 s.Under the conditions,the yield of geniposide was 71.85 ± 0.88 mg/g,(RSD = 1.233,n=3).This not onlyhas no significant difference from the predicted value 71.82 mg/g of the response surface model,but also farhigher than the yield(52.80 ± 0.88 mg/g,RSD = 1.31%,n = 3)of 50% ethanol.(2)The conversion method of gardenia blue pigment from geniposideThe conditions for converting geniposide in the extract into Gardenia blue pigment were optimized.The conversion conditions were as follows: 25 mL of gardenia extract solution contained geniposide of 1.44 mg/mL,0.4 g of cellulase addition,enzymolysis temperature of 55 ?.Under the conditions,The conversion effect of gardenia blue pigment is the best.(3)The separation of Gardenia blue pigmentThe conditions for enriching blue pigment from the conversion liquid by macroporous resin were investigated in this work.Gardenia blue pigment was adsorbed by macroporous resinHPD100 A and then eluted by 30% ethanol.The gardenia blue pigment was got with the color value of 133 and the yield of 2.5%.
