Gardenia jasminoides Ellis is a traditional Chinese medicine with rich physiological activity.The main ingredient genioside and Gardenia yellow pigment are widely used in the pharmaceutical industry,food industry and other fields.And has a huge market potential.Currently,the low purity of product is the main reason which limits the development of Gardenia jasminoides Ellis.Therefore,establishing a detection method,purification technology for Gardenia jasminoides can improve the utilization of crude medicine,that is significant to enhance the economic value of Gardenia jasminoides Ellis.In order to solve the problem of low purity of geniposide and Gardenia yellow pigment,this paper establishes an enrichment and purification technology.After purified by this technology,the content of total Crocin(Crocin-I,Crocin-II)and geniposide were increased from 1.83%,24.7% to 25.96%,89.15%.Compared with the traditional method,it is rapid,simple,and practicable with high efficiency.At the same time,HPLC method for simultaneous determination of four main components in Gardenia jasminoides Ellis was established.The chromatographic condition: Waters C18 column(4.6×150 mm,5 ?m)with gradient elution using methanol and 0.2%phosphoric acid,flow rate: 1 m L.min-1,detection wavelength: 325 nm for chlorogenic acid,238 nm for geniposide,441 nm for crocin-I,crocin-II.Becsuse of the crocin is the main active ingredient of Gardenia yellow pigment,which was also exist in saffron.It has anti-oxidant and anti-fatigue activity,so Gardenia yellow pigment could be used as a cheap substitute for saffron in-depth study,for the further development of gardenia lay the foundation.In this paper,the antioxidant activity of different extracts from Gardenia jasminoides Ellis in vitro were first studied,The results showed that Gardenia yellow pigment has the strong ability of radical scavenging and antioxidant activity in vitro.On the basis of the above-described conclusions,we explore the anti-fatigue activity of Gardenia yellow pigment by decompression chamber.Through all the above experiment,we found that Gardenia yellow pigment has significant anti-fatigue activity,could used as a natural antioxidant drug with high research value and developing potentiality.
