Objective:Baishenwang prostatitis capsule is a prescription based on the clinical experience of more than 30 years of famous traditional Chinese medicine in our province.It is mainly used to treat chronic prostatitis(drenching syndrome),with remarkable clinical effect.According to the prescription,through the research of production technology,quality standard,fingerprint and pharmacological activity,it provides experimental basis and data support for further development of drug preparation.Methods:Production process research: The anti-inflammatory activity was taken as the index to screen the four extraction process routes designed,and the single factor experiment and response surface methodology were used to optimize the extraction process parameters for the best extraction process route.Taking the best extract as the research object,the type and dosage of auxiliary materials were optimized through the specific particle forming rate,angle of repose and other inspection indexes,and finally the best production process of Baishenwang prostatitis capsule was obtained.Quality standard research: adopt modern analysis methods such as high performance liquid chromatography to establish the quality standard of Baishenwang prostatitis capsule;On this basis,in order to comprehensively and effectively control the quality standard of the preparation,the fingerprint of Baishenwang prostatitis capsule was established.Pharmacological activity research: Through the non-bacterial prostatitis and anti-inflammatory experiments in rats,as well as the anti-inflammatory,immune and analgesic experiments in mice,the pharmacodynamic effect of Baishenwang prostatitis capsule in treating chronic prostatitis was clarified.Results:In the study of production process,extraction process route 1 has obvious anti-inflammatory effect.Using single factor and response surface methodology,the bestextraction process for alcohol extraction of Phellodendron and Achyranthes bidentata was obtained as follows: 6 times of 65% ethanol for 3 times,1.6 h each time;The best water extraction technology for Portulaca oleracea L.and Astragalus membranaceus L.is 8 times the amount of water for 3 times,1.5 hour each time.The molding process of dextrin as an auxiliary material to make capsules after wet granulation was determined.In the study of quality standard,the TLC identification methods of phellodendron,Sophora flavescens,Astragalus membranaceus and saponaria vaccaria were established,and the content determination methods of berberine hydrochloride and astragaloside were established through the methodological investigation.The weight difference limit of five batches of samples was within 10%,and the whole part disintegrated within 30 minutes,and the water content was not more than 9.0%.In the study of fingerprint,the 2004 version of TCM chromatogram was used The similarity evaluation system of fingerprint has established the evaluation mode of 13 common peaks.The common peaks are classified by the comparison method of reference materials,and 6 peaks are identified.In the study of pharmacological activity,Baishenwang prostatitis capsule can inhibit the increase of leukocyte number and the decrease of lecithin body density,the decrease of TNF-?,1L-1 ?,1l-8 content,and the inhibition of foot swelling;it can inhibit the formation of agar granuloma in mice,which shows that boshenwangqianli capsule has good anti-inflammatory activity;it can improve the immunity of mice by increasing the content of hemolysin and improving the phagocytic function At the same time,boshenwangqianlie capsule can reduce the number of pain,writhing and licking feet in mice,indicating that it has a certain analgesic activity.Conclusion:In this experiment,a capsule for the treatment of chronic non-bacterial prostatitis was prepared by using modern Chinese medicine extraction and pharmaceutical technology.Using modern Chinese medicine analysis technology,the accurate and reliable quality standard and fingerprint of Baishenwang prostatitis capsule were established.Studies on pharmacological activities show that Baishenwang prostatitis capsule hasanti-inflammatory,analgesic and immunity enhancing activities,which provide strong experimental data support for its development into a pharmaceutical preparation for the treatment of chronic prostatitis.
