According to the theory of traditional Chinese medicine?TCM?coordination chemistry,the organic components,trace elements and the metal complexes formed in the process of TCM decoction may be the material basis for the efficacy of TCM.At present,the research on the medicinal ingredients of TCM mainly focuses on the separation,purification and identification of organic ingredients,or,the contents and species determination and speciation analysis of trace elements,while the research on the metal complexes in the TCM decoction is still very weak.Rutin is a common flavonoid,which is widely distributed in nature.It exhibits a variety of bioactivities,including anti-bacterial,anti-oxidation,anti-inflammatory and anti-cancer.The molecular structure of rutin contains a carbonyl group and four phenolic hydroxyl groups.Owing to these structural characteristics,rutin is easy to form complexes with metal ions.Currently,rutin complexes with various metal ions have been obtained by chemical synthesis,and studies have shown that the bioactivity of these rutin-metal complexes is significantly higher than that of rutin ligands.But these complexes are synthesized in organic solvents,which is quite different from most traditional Chinese medicines that are taken after being boiled in water.Since the study of rutin-metal complex in aqueous extracts of TCM has not been reported,this paper firstly proves that the complex formed by rutin and various metal trace elements does exist in aqueous extracts of TCM.Secondly,effects the eight kinds of metal ions on the equilibrium solubility of rutin and apparent oil-water Partition coefficient were studied.Furthermore,Under the traditional Chinese medicine extraction technology,the ability of rutin to form complexes with eight metal ions was researched and the complex was characterized structurally.Finally,the activity and possible mechanism of DNA cleavage in these complexes are understood.The main contents and results of the research are as follows:?1?Analysis of rutin-metal complexes in aqueous extracts of medicinal plants.High performance liquid chromatography?HPLC?was used to determine the methanol extracts and aqueous extracts of medicinal plants including Sophorae Flos,Lonicerae Japonicae Flos,Forsythiae Fructus,and Violae Herba,which were dissolved in methanol and 15%formic-methanol respectively.The results show that rutin does not form complexes with trace metals in methanol,but in hot water.?2?Effects the eight kinds of metal ions on the equilibrium solubility of rutin and apparent oil-water Partition coefficient.Rutin with metal ion in 8 respectively after the reaction product of solubility in the water and different p H buffer and apparent oil-water partition coefficient,different degrees of changes have taken place,possibly because the prompt metal trace elements in Chinese traditional medicine affect the organic composition in the absorption and metabolism in the body.?3?Study on the ability and structural characterization of rutin to form coordinate complexes with eight metal ions.HPLC was applied to detect the changes in the rutin content before and after the reactions of rutin with eight in hot water.The experimental result shows that rutin has the strongest ability to to form complexes with Zn2+.Besides,electrospray ionization-time of flight mass spectrometry?ESI-TOF MS?were used to detect the supernatant after the reaction between rutin and eight metal ions.The result shows that rutin could form complexes with the eight different kinds of metal ions at different ratios.?4?Study on the activity and mechanism of rutin-metal complexes on plasmid p BR 322 DNA cleavage.The results showed that the solvent DMSO,rutin,Ca2+,significant effect on DNA after reacting 24 hours.When Cu2+acted alone,it had a weak cleavage effect on DNA.Measurements of DNA cleavage activity showed that the rutin-copper complex led to dose-and time-dependent DNA cleavage through oxidative damage,which was not observed when rutin alone was reacted with DNA.In this paper,the research results show that the organic constituents in traditional Chinese medicine form complexes with trace metal elements during the water decoction process,which by increasing solubility in water,improve the absorption and enhancement activity,such as ways to improve the curative effect of traditional Chinese medicine.
