Natural plant polysaccharides are rich in resources,non-toxic,and biodegradable.In recent years,research on polysaccharide has become a focus of attention.The morphology of Elaeagnus Angustifolia Polysaccharide was characterized by electron scanning electron microscopy.The molecular weight and monosaccharide composition of Elaeagnus Angustifolia Polysaccharide was analyzed by high-performance gel permeation chromatography(HPGPC)and ion exchange chromatography(IC).Secondly,through the combination of in vitro cell experiments and animal experiments,in-depth investigations into the improvement of skin barrier function and its prevention mechanism of hyperplasia of mammary glands.results are as follows:(1)The crude polysaccharide was obtained by hot water extraction method.After deproteinization and decolorization treatment,the polysaccharide of Elaeagnus Angustifolia Polysaccharide was obtained by alcohol precipitation and drying.The average molecular weight Elaeagnus Angustifolia Polysaccharide was 2560545 Da by high performance gel permeation chromatography.The chromatographic analysis of Polysaccharides from Elaeagnus Angustifolia Polysaccharide was composed of rhamnose,arabinose,galactose,glucose,xylose,mannose,ribose,galacturonic acid,and glucuronic acid.The molar ratio of each monosaccharide is: 4.89 : 33.87 : 37.65 : 2.72 : 5.07 : 10.64 : 0.01 : 2.13 :3.02.(2)We investigated the effect of EAP on epidermal barrier function in mice model with dry skin disease and its possible underlying mechanism.Histological analysis shows that EAP can effectively reduce dry-like skin conditions,including reducing the rate of percutaneous water loss and reducing the infiltration and thickening of inflammatory cells in the epidermal layer of mice.Our research shows that topical application of EAP can inhibit various inflammatory mediators Production and increase AQP-3,FLG and LOR m RNA expression.These results indicate that topical use of EAP enhances epidermal permeability and barrier homeostasis,at least in part due to inhibition of various pro-inflammatory cytokine levels and up-regulation of AQP-3,FLG,and LOR expression..(3)To explore the protective effect of EAP on the mammary gland in vivo through the mouse mammary gland hyperplasia model induced by the combination of estradiol benzoate and progesterone.EAP can effectively reduce the diameter of mice's nipples and improve the pathological morphology of breast tissues.The serum E2 content and P content have decreased.The results show that the disturbance of serum sex hormone levels in mice has been effectively improved,and the expression of ER? and PR in the breast has been reduced.Basically down-regulated the expression of Bcl-2 and increased the expression of Bax.Therefore,EAP can effectively alleviate the estrogen and progesterone levels in mice,and effectively relieve The degree of breast hyperplasia.
