In recent years,governments interior or exterior attach great importance to the development of stem cells,immunocyte,gene therapy.etc.which promote the increasing demand for cell culture room construction.But there is no regulations or standards for the construction of cell culture room and there are few researches on airflow in cell culture room,especially on the cross-contamination caused by airflow.Therefore,through the qualitative and quantitative analysis,research was done on the airflow pattern under safety cabinet operating window in cell culture room,so as to provide some instructions for design and construction inside cell culture chamber and improve the safety of cells,personnel and environment in experimental process.Research process on this paper: Firstly,through extensive literature papers on the research topic and research methods,it is known that the most effective methods to maintain cleanness are pressure difference and airflow pattern.Secondly,field experimental data was made to obtain air velocity,pressure difference and airflow pattern in cell culture room,which was used to build and validate Airpak model of airflow in cell culture room.Thirdly different scenarios were simulated including biological safety cabinet circulation air ratio,biological safety cabinet air velocity,safety cabinet window opening height,indoor air volume and indoor opening location.etc.to figure out how they influence air pressure difference and flow pattern under the operating window.Finally research and analysis were made on flow optimization under operating window by adding a wind curtain inside safety cabinet or return openings surrounding operating window.Conclusions are drawn as follow:(1)By comparing and analyzing the experimental data and simulating data,Airpak simulation of cell culture room airflow organization model has great accuracy;(2)With no operator in front of the safety cabinet mimicking reality,no crosscontamination occur between the two spaces under different designing factors but there is leak of air along operating arms when person is present;(3)percentage of safety cabinet exhaust air and velocity of its air supply have great influence on pressure difference and flow pattern under operating window between safety cabinet and cell culture room.With a constant air supply of 0.4m/s in safety cabinet,exhaust air ratio of no less than 30% or with 30% exhaust ratio,wind speed in biological safety cabinet no higher than 0.6m/s,is able to have a better control of air flow pattern in and out the cabinet,and pressure from inside the biological safety cabinet to the outside should be less than-0.23 Pa,or lumen overflow will occur;4)By adding exhaust opening around the operating window,the running out of cabinet air along the steady operating arm can be effectively put under control.
