Moringa oleifera Lam.,also known as the Drumstick tree,is a perennial tropical deciduous tree growing the northern Indian.Currently,this plant is also cultivated in the areas of Hainan and Yunnan provinces in China.Totally,there are 14 species of this genus around the world.Notably,the seeds of M.oleifera possess varities of biological activities such as antihyperlipidemic,antiaging and immunoregulating actions,and widely used in the folk.The aim of this thesis is to explore the immunoregulating active compounds in M.oleifera.In the first,the seeds of M.oleifera was subjected to extract withm ethanol,solid phase extraction with petroleum ether,chloroform,ethyl acetate and methanol,and further purified with column chromatographies including macroporous assorption resin,silica gel,Sephedex LH-20 colums,and preparative high performance liquid chromatography.Finally,there are 11 compound isolated form the seeds of M.oleifera.After spectroscopic elucidation with 1H-NMR?13C-NMR?1H-1H COSY?HMBC?HSQC?NOESY and ESI-MS,these 11compounds were identified as glucosinalbin?1?,Moringa A?2?,Phenyl-methanoid?4-hydroxyphenylethyl4-O-?-D-glucopyranosyl-?1?3?-O-?-L-rhamnopyranoside??3?,4'-O-??-L-rhamnopyranosyl?glucosinalbin?4?,benzylalcohol-?-vicianoside?5?,p-hydroxybenzoic acid 4-O-?-D-glucopyranosyl-?1?3?-?-L-rhamnopyranoside?6?,Uridine?7?,adenosine?8?,Marumoside A?9?,4-Hydroxybenzaldehyde rhamnoside?10?,vitexin?11?.Among these isolated compounds,there are one carbamate ester,three phenylethanoid glycosides,four phenolic glucosides,two nucleosides and one flavonoid Glycoside.And compond 2 and 5 are new compounds,and compound 3,7,8 and 11 are the first time isolated in this plant.In vitro,we also evaluate the immunoregulating action of these isolated compounds on the release of endogenous antiviral factors such as TNF-??IFN-??IFN-??IL-1?IL-2?IL-6?IL-12?IL-15?IL-18?IL-21 from macrophage Raw264.7 cells and found that glucosinalbin?compound 1?could significantly upregulate the release of TNF-?from macrophage Raw264.7 cells.Take it together,This work enriched the chemical component of the seeds of M.oleifera and revealed its immunoregulating active component,which laid the foundation for the development and pharmacological use of M.oleifera.
