Objective:Rose crown chicken is unique in its delicate fleshy texture,and its huge mulberry like crown shape is eye catching.It is a valuable breeding material for poultry and is expected to be cultivated into high quality Xinjiang local breeds(supporting chickens).In this study,rose crown chicken as material,Liangfeng chicken as a reference to explore the genetic characteristics of genes affecting muscle quality,looking for molecular markers can be used for assisted breeding of rose crown chicken,providing a reference basis for genetic improvement of rose crown chicken.Methods:The crown traits of rose crown were identified by DNA molecular marker method.The contents of AMP,IMP,and HYP in muscle tissue were determined by high performance liquid chromatography.The real-time fluorescence quantitative PCR was used to explore H-FABP,A-FABP and ADSL genes in it.At the time of growth and development,the SSCP technique was used to detect the SNPs of H-FABP,A-FABP and ADSL genes,and correlated with the muscle quality of rose crown chickens.Results:(1)Genotypic molecular identification was show that three genotypes RR,RS,and SS were found in Rose Crown Traits and single-crown traits,the evaluation molecular markers of results by traditional method testcross,F1 generation crown of results show that the results of testcross are completely consistent with molecular identification.(2)Routine nutritional of muscles analysis that chest,leg muscles of rose crown chickens,whose IMF were significantly higher than Liang feng chickens(P<0.05).The taste substances measurement showed that,which Liang feng chicken’s breast muscles AMP was significantly higher than roses chicken’s pectoral and leg muscles(P<0.01),IMP content of chest muscles was highest in rose crown,it was significantly higher whose leg muscles as well as Liang feng chicken’s pectoral and leg muscles(P<0.01),the crown chicken’s HYP content in leg muscles was significantly higher than their chest muscles as well as Liang feng chicken’s breast muscles(P<0.01),showing significant differences in cultivars.(3)The results of qRT-PCR showed that the mRNA expression of H-FABP gene decreased with the increase of the age,and negatively correlated with the IMF content of the breast and leg muscles of the two breeds,but did not reach a significant level;A-FABP showed an upward trend.And there was a significant positive correlation between the IMF content of the breast and leg muscles of the two breeds of chickens;the expression of the ADSL gene showed a fluctuating change,both showing a first rise and then a decrease,and the final expression rose back to a higher level.There is a significant positive correlation between IMP content in muscle.(4)SSCP technology detects H-FABP(T-632C,G-747A,G-896A),A-FABP(A-1765G,G-1978A),ADSL(A-4677G,T-5611A,C-8335T),T-15008C,G-15484A)SNPs,H-FABP,A-FABP genes and IMF content analysis showed that:T-632C,G-896A,A-1765G SNP loci on the breast muscle of two breeds of chickens,Leg muscle IMF content had a significant effect,AB and BB type were the dominant genotypes of this site.After combining the favorable genotypes of the two genes,ABBB polymorphism had the highest IMF content in leg muscles and was significantly higher than AAAA,AAAB,BBAA individuals(P<0.05).Conclusion:Molecular identification method can be used to replace the traditional test-crossing method for the selection of crown-type traits in rose crowned chickens.The muscle quality of rose crown chickens is better than that of Liangfeng chickens,and the expression of A-FABP,ADSL gene and muscle IMF,IMP The content was significantly positively correlated;T-632C,G-896A,and A-1765G polymorphisms were significantly associated with the IMF content of rose-crown muscles.The predominance of the ABFB genotype polymerized individuals had the highest IMF content in the leg muscles.These sites were used as molecular markers related to muscle quality traits of rose-crown chickens,and the selection of rose-crown chickens was used.
