| [Objective] Using the albumin and glutenin from pleurotus eryngii as raw materials,the research is mainly to obtain antioxidant peptides by enzymatic hydrolysis,and explore its intestinal absorption capacity,they could provide a theoretical basis for the development of pleurotus eryngii polypeptide products.[Methods](1)Using the hydrolysis degree,the scavenging rate of O2-·,·OH,DPPH·and total reducing power as indexes,the optimum enzymes for hydrolyzing Pleurotus eryngii albumin and gluten were selected from seven proteases(alkaline protease,papain,pineapple flavor protease,protease,neutral protease,trypsin,acid protease).(2)Sephadex G-25 gel filtration chromatography was used to purify the antioxidant peptide from Pleurotus eryngii,and the purified polypeptide solution was collected.The molecular weight range of antioxidant peptide from Pleurotus eryngii was determined by time-of-flight mass spectrometry.(3)The absorption capacity of antioxidant peptides from pleurotus eryngii in different intestinal segments of rats was studied by everted intestinal sac model.[Results](1)The optimum enzymes for hydrolyzing albumin of Pleurotus eryngii are neutral protease and papain combined enzymes,the scavenging rate of O2-·,·OH,DPPH·and total reducing power are79.79%,75.44%,78.79% and 0.209.The optimum enzyme for hydrolyzing glutenin of Pleurotus eryngii are neutral protease and papain combined enzymes,the scavenging rate of O2-·,·OH,DPPH·and total reducing power are 72.32%,73.91%,77.13% and 0.219.Polypeptide of Pleurotus eryngii with certain antioxidant activity was prepared.(2)The molecular weight of the antioxidant peptides of albumin and glutenin in pleurotus eryngii are104~764Da and 203~820Da,both of which are small molecular polypeptides.(3)The antioxidant peptides of albumin and glutenin are absorb in different parts of the small intestine of rats.Compared with duodenum and ileum,Compared with duodenum and ileum,the absorption of antioxidant peptide of albumin and gluten in jejunum is stronger.[Conclusion] The antioxidant peptide of Pleurotus eryngii was prepared,and it can be directlydigested and absorbed in the small intestine.Therefore,it can provide theoretical basis for the development of polypeptide related products of Pleurotus eryngii. |
