Gastrodia elata has a history of more than 2000 years in China,and its safety and effectiveness have been fully proved by a large number of clinical applications.Modern studies have shown that Gastrodia elata has an effective effect on the central nervous system,cardiovascular system and immune system,mainly contains phenols and its glycosides,organic acids,polysaccharides and other effective components.In this paper,two new breeding materials of Sichuan Tianma Nanwu No.1(TMNW-1)and Sichuan Tianma Ahong No.1(TMAH-1)were used as the main experimental materials,and different germplasm resources from other regions of Sichuan Gastrodia elata were used as contrast materials.referencing to the Chinese Pharmacopoeia(2015 edition)and domestic and foreign literature,the contents of moisture,ash,leach,polysaccharide,gastrodin and p-hydroxybenzyl alcohol in Gastrodia elata were determined,and the quality was evaluated comprehensively according to the results of the detection indexes of Gastrodia elata..The antioxidant activity of Gastrodia elata in vitro was studied by salicylic acid method,FRAP method and DNA oxidative damage protection method and the HPLC fingerprints of different germplasm resources of Gastrodia elata from different habitats were established based on the optimization of reflux extraction technology by response surface method.The relationship of its antioxidation activity in vitro was studied by the combination of fingerprint and antioxidant activity.The qualitative and quantitative models of 25 batches of samples from Jiuzhaigou(including two new materials),Jinkouhe and Jiangyou were established.The quality standard and quality control technical system of two kinds of new materials of Sichuan Gastrodia elata were drafted.The main findings are as follows:1.The moisture content of two new materials and control samples was determined by drying method.The moisture content of two new materials and control samples was 7.15%,and the average content was 8.41%;The total ash content was 2.10~3.80%,the average content was 2.92%,the content of acid insoluble ash was 0.16%~0.46%,the average content was 0.31%,according to the Chinese Pharmacopoeia(2015 edition,No.1);The contents of alcohol extract and water extract were 12.87~26.62%、21.73~49.59%,and the average contents were 20.53% and 34.46%,respectively.The content of polysaccharide was between 10.75% and 29.2%,determined by sulfuric acid-anthrone method;the sum of gastrodin and p-hydroxybenzyl alcohol was 0.2953% and 0.7017%,the average value was 0.419%.The comprehensive score of Sichuan Tianma Nanwu No.1(TMNW-1)and Sichuan Tianma Ahong No.1(TMAH-1)were the first and the third,respectively.2.The response surface optimization method was used to optimize the reflux extraction process of antioxidation components from Gastrodia elata.The optimum extraction process was as follows: material-liquid ratio of 1: 55,extraction temperature of 65 ℃,extraction time of 66 min.3.The results of hydroxyl radical scavenging rate were as follows: the IC50 value of hydroxyl radical scavenging rate of new material TMNW-1 was the highest,that of TMGH was the lowest,and that of new material TMAH-1 was the second;The results showed that the total antioxidant capacity of the two new materials was not good,and the two new materials could inhibit the oxidative damage of DNA to a certain extent,but they did not perform well in all the samples to be tested.4.The HPLC fingerprints of two new materials and other 8 samples were established and 15 common peaks were calibrated.The range of similarity is 0.923 to 0.997.Similarity and cluster analysis results are consistent,10 samples are divided into two categories5.The relationship between fingerprint and antioxidant activity of Gastrodia elata was studied.The results showed that there were 6 antioxidant peaks related to hydroxyl radical scavenging rate and 7 antioxidation peaks related to total antioxidant capacity.6.By using discriminant analysis(DA),25 samples of Gastrodia elata,including two new materials,were successfully identified,the correct rate was 100%,and the infrared prediction model of total contents of gastrodin and p-hydroxybenzol was established.7.The quality standard(draft)and quality control technology system of new breeding materials TMNW-1 and TMAH-1 were drawn up.In this paper,the quality and antioxidant activity of two new materials and different germplasm resources of Gastrodia elata from different habitats were studied,and the fingerprint and infrared model of total content of gastrodin and p-hydroxybenzol were established.It provides a scientific basis for the quality control and application development of two new materials and other samples of Gastrodia elata.
