Objectives Serum glucose and vaspin levels were measured in preganant women between 24 and 28 weeks gestation after a 75 g oral glucose challenge in order to explore the correlation between serum vaspin levels and glucose levels as well as that between serum vaspin and gestational diabetes mellitus(GDM).Methods A total of 60 cases of singleton pregnancy,including pregnant women with GDM(n = 30)and pregnant women without GDM(n = 30),were recruited from Hebei General Hospital Obstetrics and Gynecology from November 2016 to April 2017.All women were between 24 and 28 weeks gestation.Record the age of pregnant women,gestational age,pre-pregnancy weight,the current height and weight.The steady-state model was used to evaluate the insulin resistance index(HOMA-IR),the pre-pregnancy body mass index(BMI-1)and the current body mass index(BMI-2).Glucose oxidase was used to detect two groups of fasting blood glucose(FPG),1h postprandial glucose(1h-PG)and 2h postprandial glucose(2h-PG)levels.Using automatic biochemical analyzer to determine lipid levels,blood lipids including total cholesterol(TC),triglycerides(TG),high-density lipoprotein(HDL),and low density lipoprotein(LDL);Two groups of fasting insulin(FINS)were detected by radioimmunoassay.The enzyme linked immunosorbent assay(ELISA)was used to determine two groups of serum vaspin,1h-vaspin and 2h-vaspin levels.Results 1 FINS?HOMA-IR?FPG?1h-PG?2h-PG?oh-vaspin?1h-vaspin?2hvaspin levels in the GDM group[(15.08±6.30)?U/mL,(3.47±1.56),(5.09±0.47)mmol/L,(9.57±1.47)mmol/L,(7.88±1.30)mmol/L,(0.77±0.39)ng/mL,(1.04±0.42)ng/mL,(0.97±0.41)ng/mL] were significantly higher than those with NGT[(11.42±5.32)?U/mL,(2.32±1.13),(4.53±0.27)mmol/L,(7.67±1.04)mmol/L,(6.61±0.86)mmol/L,(0.50±0.32)ng/mL,(0.60±0.31)ng/mL,(0.56±0.28)ng/mL],respectively(P<0.05);2 1h-PG and 2h-PG were significantly higher than FPG in both groups(P<0.05);1h-PG were significantly higher than 2h-PG in both groups,(P<0.05);Blood glucose levels in women with gestational diabetes mellitus(GDM)increased to more extent than in normal pregnancy,including d1(1hPG-FPG)? d2(1h-PG-2h-PG)and d3(2h-PG-FPG)[(4.48 ±1.49)?(1.70±1.25)?(2.78±1.30)mmol/L]vs[(3.14 ±1.01)?(1.06±1.12)?(2.08±0.88)mmol/L],respectively(P <0.05);3 0h-vaspin ?1h-vaspin and 2hvaspin levels in women with GDM[(0.77±0.39)?(1.04±0.42)?(0.97±0.41)ng/mL] were significantly higher than those with NGT[(0.50±0.32)?(0.60±0.31)?(0.56±0.28)ng/mL],respectively(P <0.05);D1(1h-vaspin-0hvaspin)were significantly higher than those with NGT[0.19(0.12,0.36)vs 0.09(0.02,0.25)ng/mL],(P <0.05).4 2h-vaspin had a positive correlation with 1h glucose in women with NGT(r= 0.384,P= 0.036);0h-vaspin?1h-vaspin and 2h-vaspin had a positive correlation with 1h glucose [(r=0.494,P=0.006)?(r=0.577,P=0.001)?(r=0.557,P=0.001)]and TG[(r= 0.385,P=0.036)?(r=0.466,P=0.010)?(r= 0.488,P=0.006)] in women with GDM;5 0h-vaspin had a positive correlation with 1h-PG(r=0.532,P=0.000)? 2h-PG(r=0.352,P=0.006)and TG(r=0.335,P=0.009)in all women;1h-vaspin had a positive correlation with 1h-PG(r=0.651,P=0.000)? 2h-PG(r=0.450,P=0.000)? TG(r= 0.379,P=0.003)? FINS(r=0.288,P=0.026)and HOMA-IR(r=0.319,P=0.013)in all women;2h-vaspin had a positive correlation with 1h-PG(r=0.651,P=0.000)? 2h-PG(r=0.427,P=0.001)?TG(r=0.382,P=0.003)?FINS(r=0.292,P=0.024)and(r= 0.327,P= 0.011)in all women.Conclusions Vaspin is regulated by blood glucose load.Serum vaspin level may be related to the level of insulin resistance and insulin secretion decline in early phase as a compensatory secretion resulting from IR state.There is a relationship between vaspin and regulation of blood glucose as well as lipid metabolism.There is a relationship between vaspin and GDM.
