Objective : To study the correlation between FOXO1 overexpression and the autophagy behavior in vulva squamous carcinoma cell line A431,and to provide some molecular biological evidences for the study of the occurrence and development of vulvar squamous carcinoma related mechanisms.Methods:According to the GeneBank middleman FOXO1 CDS sequence by Shanghai JiKaiJiYin due to the design and synthesis of building GV144-FOXO1 eukaryotic expression plasmid analysis and appraisal,the appraisal after the transient transfection expression vector of vulvar squamous cancer cells A431,measuring FOXO1 after transfection expression plasmid transient transfection samples by immunofluorescence observations FOXO1 expression vector into cells express,western blot method(western blot)detection after transfection FOXO1,LC3 A,LC3B protein expression.Results:GV144-FOXO1 overexpression plasmid,GV144 negative control plasmid transfection into A431 cells,GV144-FOXO1 overexpression plasmid transfection group and GV144 negative control group can be obviously green fluorescent plasmid of immunofluorescence,showed that expression plasmid have been transfected into cells.In Western blot experiment,the protein expression in the GV144-FOXO1 overexpression plasmid transfection group was significantly higher than GV144 negative control group.FOXO1 transfection group of FOXO1 protein expression quantity increased obviously compared with negative control group,the difference was statistically significant(P < 0.05),and between the negative control group and blank group,FOXO1 protein expression has no significant difference(P > 0.05).Western blot detect LC3 B protein expression after transfection experimental group than the control group significantly increased,LC3A/B is reduced,.Conclusion:The overexpression of FOXO1 in the cell line A431 of vulva squamous cell carcinoma was able to enhance autophagy activity.
