| Objective: Ulcerative colitis(UC)is a chronic and non-specific inflammatory disease of the rectum and colon that is not yet clear.The disease duration and severityvary,repeatedly.In this study,we observed the effects of Wenglianjiedu decoction on the general condition,disease activity index morphology(DAI),histopathologica,l Interleukin-13(IL-13)and Toll-like receptor 4(TLR4)to explore the mechanism of Wenglianjiedu decoction in the treatment of ulcerative colitis,and to provide experimental basis for clinical application.Methods:60 healthy male Wistar rats(weight 220g-250g)fed for 7 days,were randomly divided into 6 groups,including control group(CK group),model group(MG group),mesalazine group(MS group),high dose of Chinese medicine group(HD group),middle dose group(MD group)and low dose group(LD group),10 rats in each group received routine feeding.In addition to CK group,the rats in the other groups were designed with TNBS/ ethanol combined with UC model.On the fourth day after successful modeling,the CK group was treated without special treatment.MG group was given 4ml /kg normal saline,MS group was given the mixture of mesalazine suspension,Chinese traditional medicine group rats respectively to the corresponding dose of the drug lavage,continuous drug intervention changes during the treatment of mental state,food intake,body weight of rats and the stool generally observed after 2 weeks,and DAI score.After the treatment,the rats in each group were fasted for 24 hours,blood was taken from femoral artery after lidocaine anesthesia.The levels of serum IL-13 were detected by enzyme-linked immunosorbent assay(ELISA).The expression of TLR4 in intestinal mucosa was detected by immunohistochemistry.Results:1 General state All rats in the CK group were in good spirits in this entire treatment period,like food,shiny hair neat,agile,weight gain,normal black granular stool.After successful modeling,the remaining groups of rats appeared loose stools or mucopurulent bloody stools in different degrees,perianal contamination,do not like activities,poor spirit,messy hair,eat less.Except for rats in the MG group,the other four rats gradually improved after drug intervention in mental status,appetite,weight,and then loose stools and mucus purulent blood stools decreased than before.The symptom of Rats in the HD group improved obviously,there was no significant difference between MS group and MD group,LD groups' rats were less than those three groups.With the progress of the experiment,MG groups' rats were getting worse and worse,Loose stools or mucus purulent blood stools aggravated,weight loss,depression,nuzzles,hair messy,anorexia.2 DAI score results HD group,MD group,LD group and MS group values were lower than the MG group,the difference was statistically significant(P < 0.05);MD and MS group were lower than that of LD group,the difference was statistically significant(P < 0.05);there was no significant difference between MD group and MS group(P > 0.05).3 histopathological observation of colon The surface epithelium of intestinal tissue of rats in group CK were complete,crypt long and straight,no branches,more goblet cells,there was a small amount of inflammatory cells in the lamina propria,and no thickening of the muscularis mucosae;In group MG,inflammatory exudate and necrotic tissue were seen on the surface of ulcer,a large number of inflammatory cells infiltrated in the lamina propria;In group HD,the surface of intestinal tissue was complete,the crypt was long and straight,a few branches,a small amount of inflammatory cells were found in the lamina propria;MD group and MS group was not significantly different,the surface epithelium was irregular,fewer crypts,more goblet cells,fewer inflammatory cells were seen in thelamina propria;Group LD irregular surface epithelium,crypt was branched and twisted,the number of goblet cells was significantly reduced,the distance between the crypt base and the muscularis mucosae increases,massive inflammatory cell infiltration,thickening of mucous membrane.4 Index detection The expression of IL-13: The expression of IL-13 in CK group was higher than that in HD group,MD group,LD group,MS group and MG group,the difference was statistically significant(P < 0.05);The HD group was higher than that of MD group,LD group,MS group and MG group,the difference was statistically significant(P<0.05);MD group and MS group were higher than those in LD group and MG group,the difference was statistically significant(P<0.05);LD group was higher than MG group,the difference was statistically significant(P<0.05);MD group compared with MS group,no statistical significance(P>0.05).The expression of TLR4: The expression of TLR4 in CK group was lower than that in HD group,MD group,LD group,MS group and MG group,the difference was statistically significant(P <0.05);HD group was lower than MD group,LD group,MS group and MG group,the difference was statistically significant(P<0.05);MD group,MS group was lower than LD group,MG group,the difference was statistically significant(P<0.05);LD group was lower than MG group,the difference was statistically significant(P<0.05);MD group and MS group was no significant difference(P> 0.05).Conclusion: Wenglianjiedu decoction can significantly improve the ulcerative colitis rats' general situation and Injured intestinal mucosa,the effect of high dose group was better than that of other groups,there was no significant difference between the middle dose group and mesalazine group,the effect of low-dose group was lower than that of high dose group,middle dose group and mesalazine group.Its mechanism may be by reducing intestinal tissue TLR4 expression,increasing the content of IL-13,by influencing the some kind of signal transduction pathways,increased the release of inflammatory factor suppression,to promote proinflammatoryfactors and anti-inflammatory factors tend to dynamic balance,thus blocking the process of cell damage,stimulate local tissue repair and regeneration,promote ulcer healing. |
