| Objective:The present study is to observe the effect of Danzhi compound recipe functioning as smoothing liver and relieving depression,strengthening spleen and harmonizing ying,and clearing away heat retention on thyroid function,liver function,the level of oxidative stress enzymes and pathological morphology of thyroid and liver in BALB/c female rats with hyperthyroidism,investigate the correlation between hyperthyroidism and hepatic function damage,research the correlation between hepatic lesion rats with hyperthyroidism and oxidative stress reaction.Our study aims at exploring the functioning mechanism of Danzhi co mpound recipe in preventing and treating hyperthyroidism with hepatic lesion,which provides theoretical basis for the treatment of hyperthyroidism with hepatic lesion by traditional Chinese medicine.Methods:200 healthy BALB/c female rats at the age of six weeks,weighing about 18±2g,were fed for one week in an adaptive breeding way.Randomly select ing 170 rats enrolled into model group,the remaining 30 rats as control group.The model group received adenovirus Ad CMV-TSHR289 injection at the drops of 1*108PFU/ml every three weeks and continuously immuned three times.The control group did not receive special treatment.After six weeks,the successfully established 150 modeling rats were divided into sub-group:the model group,and the group with methimazole,and the group of low dosage of Danzhi compoun d recipe,and the group of middle dosage of Danzhi compound recipe,and the group of high dosage of Danzhi compound recipe.At the same time,the model group and the control group were given distilled water,other groups respectively were given corresponding drugs by gavage.After the experiment,collected blood by picking eyeball,then detected TSH,T3,T4 with enzyme linked immunosorbe nt(ELISA);detected ALT,AST,MDA,SOD,CAT with biochemical method.Taking th yroid tissue and liver tissue stained by HE were observed for morphological.Results:1.After six weeks of treatment,TSH of model group was significantly lower than the control group,T3,T4 was higher than the control group.The western medicine group had the best effect on improve thyroid function among each treatment group.TSH,T3,T4 were restored to normal levels.Danzhi compound decoction can improve thyroid function in a certain extent,but can?t reduce it to normal levels.2.After six weeks of treatment,AST,ALT,ALP of model group were significantly higher than control group.Danzhi compound decoction can significantly improve liver function,especially in the group of middle dosage of Danzhi compound decoction.The western medicine group still suffer hepatic lesion in different degree.3.After six weeks of treatment,MDA,SOD,CAT of model group was significantly higher than control group.Danzhi compound decoction can reduce oxidative stress reaction significantly.The western medicine group still suffer oxidative stress damage.4.Hepatic lesion and oxidative stress damage were positively correlated with hyperthyroidism.Oxidative stress damage was positively correlated with hepatic lesion.Conclusion:1.Immuned by adenovirus AdCMV-TSHR289 can successfully create the model of hepatic lesion with hyperthyroidism.Methimazole and Danzhi compound decoction can treat hyperthyroidism,and the effect of methimazole may be better than the Danzhi compound on the recovery of thyroid function.2.The hepatic lesion is positively correlated with hyperthyroidism.Methimazole and Danzhi compound decoction can improve liver function,and the moderate doses of Danzhi compound decoction is the most significant therapeutic dosage.3.The level of oxidative stress reaction is positively correlated with hyperthyroidism and hepatic lesion.Methimazole and Danzhi compound decoction can effectively reduce oxidative stress damage.4.This experiment shows that there is no hepatic toxicity in each dosage of Danzhi compound decoction,and it can effectively improve hepatic lesion,we can further research and apply in clinical treatment. |
