| Objective:The present meta-analysis was conducted to make an estimate in efficacy and safety of dapoxetine in men with premature ejaculation.Methods:We searched Embase,PubMed,Cochrane,Web of Knowledge,FDA.gov,and Clinical Trials.gov systematically for studies reporting dapoxetine in men with premature ejaculation.Our main endpoints were intravaginal ejaculatory latency times(IELT),personal distress related to ejaculation(PDRE),and treatment-emergent adverse events(TEAEs).We assessed pooled data by using a random-effects model with STATA 12.0 software.It has been registered at International Prospective Register of Systematic Reviews(PROSPERO)(number CRD42017065186).Of 276 identified studies,a total of 8 studies were selected in our analysis.Results:Compared with placebo,dapoxetine was significantly efficacious in men with PE.The result indicated that patients who were treated in dapoxetine could benefit from physiology(IELT overall: WMD = 1.67,95%CI:1.45-1.89)and psychology(PDRE overall: RR = 1.26,95%CI: 1.18-1.35).We also assessed the difference of 3 treatment strategies: 30 mg ondemand use(WMD = 1.38,95%: 1.01-1.75),dapoxetine 60 mg on-demand use(WMD = 1.62,95%: 1.40-1.84),dapoxetine 60 mg daily use(WMD = 2.18,95%: 1.71-2.64)firstly.Most TEAEs were associated with the treatment and drug dose,and overall safety of dapoxetine was acceptable.Conclusion:In conclusion,this review and meta-analysis provides clear evidence of the benefits of dapoxetine in men with premature ejaculation,including multiple doses and treatment strategies,however we still need more RCTs from neutral institutions in the future and more further studies of dapoxetine in different strategies should be performed to application of this treatment in practice. |
