A Meta-analysis And Systematic Review Of Dapoxetine For Treatment Of Premature Ejaculation

Objective To assess the efficacy and safety of Dapoxetine for treatment of premature ejaculation and provide the evidence for clinical decision-making. The involved studies need to be analyzed is their intravaginal ejaculatory latency time( IELT),patient-reported global impression of change( PGI), and composite PRO criteria for clinical benefit(CCCB),including satisfaction with sexual inter-course(SWSI), perceived control over ejaculation(PCOE) and personal distress related to ejaculation(PDRE).Methods We searched PubMed, Embase, BIOSIS Previews, Cochrane Library,CNKI Database and Wanfang Database for literature on Dapoxetine for treatment of premature ejaculation. We performed meta-analysis on the identified publications and evaluated its therapeutic efficacy and safety that based on the collected studies.Results Six relevant randomized controlled trials(RCT) were included which involving about 12658 cases of premature ejaculation. Compared with placebo controls,the patients treated with Dapoxetine had showed significant improvement in IELT[WMD= 0.48,95%CI(0.28,0.68),P=0.0010],PGI[RR=1.17,95%CI(1.09,1.25),P=0.01],SWSI [RR= 1.11,95%CI(1.06,1.16),P<0.00001],PDRE[RR=1.26,95%CI(1.19,1.34),P<0.00001] and PCOE[RR=1.16,95%CI(1.08,1.24),P<0.00001].Meanwhile, there were significant differences between the 60 mg and 30 mg Dapoxetine groups in IELT[WMD=0.48,95%CI(0.28,0.68),P=0.0010],PGI[RR=1.17,95%CI(1.09,1.25),P=0.01],SWSI[RR=1.11,95%CI(1.06,1.16),P<0.00001],PDRE[RR=1.26,95%CI(1.19,1.34),P<0.00001],PCOE[RR = 1.16,95%CI(1.08,1.24),P<0.00001]as well.Conclusion The available conclusion indicated that no matter 60 mg or 30 mg Dapoxetine can significantly prolong IELT and greatly improved CCCB(PGI, SWSI,PDRE and PCOE), and has a better effect on 60 mg dose usage indeed.
