Cynomorium songaricum Rupr has been titled as "elixir",which is an obligate root parasitic plant.It is mainly distributed in Northwestern China and Central Asia,where are dry and rocky.It has been used as a folk medicine for reinforcing kidneys,benefiting the anima and moisturizing the intestine based on the warm sweet.The objective of the study was to screen the antioxidant from Cynomorium songaricum Rupr.It mainly dealt with the extracting technology,screening the fraction of antioxidant activity and screening the antioxidant compounds in the study just as follows:1 Extraction time,the concentration of solvent and ratio of liquid-solid were chosen as independent variables.The antioxidant capacity determined by the CUPRAC was employed the dependent variable.The optimized extracting technology for the total antioxidant compounds from Cynomorium songaricum Rupr was determined by design-response surface methodology.The optimized extracting technology of Cynomorium songaricum Rupr was selected as follows:the ratio of liquid-solid was 13:1,the concentration of alcohol was 50%and processing time was 1.5 h.CUPRAC with design-response surface methodology,which was used to optimize the extracting technology for antioxidant compounds from Cynomorium songaricum Rupr,was more reliable.2 The antioxidant activity of different fraction of Cynomorium songaricum Rup was studied in vivo and vitro essays.Several fractions of Cynomorium songaricum Rup extracted by petroleum ether,ethyl acetate,n-butyl alcohol and water were determined by CUPRAC in vitro essays.It was found that the fraction of Cynomorium songaricum Rup extracted by ethyl acetate possessed the highest antioxidant activity,which was selected as the fraction of antioxidant activity.The antioxidant effect of different fraction from Cynomorium songaricum Rupr on the subacute mice aging induced by injection of D-galactose was studied.The biological redox substance,such as SOD,MDA and GSH-Px,in the brain,liver and kidney were determined.To evaluate the antioxidant capacity for different extraction in vivo assays.The results showed that the best fraction of Cynomorium songaricum Rupr in vitro essays was ethyl acetate.3 The principle for screening antioxidant compound based on the reaction between Cu(II)-Nc with compound,which would show yellow results.TLC-autography combined with Silica gal column was applied to screen the antioxidant compounds from the fraction of ethyl acetate.Five antioxidant compounds were isolated and identified by 1H-NMR,13C-NMR,and ESI-MS as dapilliplactone(1),protocatechuic acid(2),catechin(3),gallic acid(4)and epicatechin gallate(5).Besides,stearic acid a-monoglyceride(6),?-sitosterol(7),n-butyric acid(8),daucosterol(9)were isolated from the fraction of ethyl acetate of Cynomorium songaricum Rupr at the same time.
