| Procyanidin is a type of polyphenols substance which has been continuously researched and developed as a strong active functional factor recently and widely exixts in nature plants and food fields. A mass of researches indicate that procyanidins has many outstanding characteristics such as strong antioxidant capacity, ability of elimininating free radicals and better pharmacological activities.Today, procyanidins has been widely used in many fields such as the healthy food, medicine and cosmetics etc.In the field of procyanidin,grape seed is an everlasting study subject.Therefore taking full adventage of the abundant plant resources, especially herba medicine resources in our country to extract the natural antioxidant procyanidins is of great significance.As a kind of Chinese herba medicine Cynomorium songaricum not only contains flavonoids, nonsteroidal, triterpene ,but also minerals, amino acids.Much emphasis has been put on studies of Cynomorium songaricum .However no report has been found on the research of the procyanidins of Cynomorium songaricum at present. This project aims to find more nature plant to replace the sole grape resources.On the other hand ,we hope to further expand the value of Cynomorium songaricum through development and utilization of procyanidins.This thesis mainly includes three parts:(1) Procyanidin is found in Cynomorium songaricum extract were systematically analyzed by reversed phase liquid chromatography-electrospray mass spectrometry (RP-HPLC/ESI-MS) and corresponding molecular ions [M-H] of ESI- ionization procyanidin monomer and oligomers were identified. As a result, two proanthocyanidins monomer, six proanthocyanidins dimer, as well as five Proanthocyanidins trimer were detected from Cynomorium songaricum extract, while the polymers of gallic acid ester were not found. The result indicates that procyanidin can be found in Cynomorium songaricum and the corresponding procyanidins are non-ester procyanidins.(2)The extraction of Cynomorium songaricum was studied. The dried power of Cynomorium songaricum was used as raw material. Five extraction factors were studied by orthogonal experiment, such as concentration of solvent, extraction temperature, solution pH, ratio of solvent to Cynomorium songaricum power, extraction times. Comparing with the results of orthogonal experiment, the optimal extraction conditions were as follows: ultrasonic extraction for 2h, 80% ethanol, 40℃, pH 5,ratio of solvent to Cynomorium songaricum power was 8:1. Proanthocyanidins has been quantitatively detected in Cynomorium songaricum that obtained from different places. As a result, Cynomorium songaricum produced in Inner Mongolia Alxa with higher procyanidins, reaching 13.42%. However, it may due to the impact of the growth environmental, such as climate and soil conditions, Cynomorium songaricum product in Gansu Jiuquan contents lower procyanidin than the other three Cynomorium songaricum materials.(3) The antioxidant activities of proanthocyanidins from Cynomorium songaricum and grape were studied on ability of elimininating DPPH radical.With the increase of extract concentration, the capacity of Cynomorium songaricum of elimininating DPPH radical increased. The rate can reach to 81% and slightly higher than the grape seed extractions with a rate of 79%.. Therefore, Cynomorium songaricum not only has effect on balance of feminine and masculine, but also may have good developing prospects as effective free radical scavenger. |
