| ObjectiveGanderma lucidum(Leyss.ex Fr.)Karst,also known as Wannianrong or Mulingzhi or Lingzhicao,which belongs to the family of Ganoderma genus,Polyporaceae.Ganoderma lucidum has been used by pharmaceutical fungi in folk for thousands of years,with very high medicinal and health value.Ganoderma lucidum spore is the fungus's reproductive cell collecting from the cap of Ganoderma lucidum during the time of maturity,with most of the inherited active ingredients of Ganoderma lucidum.There are triterpenes,sterol,amino acid,polysaccharides alkaloids and many other functional compositions in Ganoderma lucidum.Clinical pharmacology researches have proved that the spore was active in resisting tumor and improving immunity,which is better than Ganoderma lucidum.Ganoderma lucidum spore oil is a mixture oil that is extracting from Ganoderma lucidum spore by supercritical carbon dioxide extractive technology.There are vegetable fatty acid triglycerides,free fatty acids and a small amount of triterpenes,sterol in Ganoderma lucidum spore oil.Modern pharmacology researches show Ganoderma lucidum spore oil can resistive tumor,adjust immunity,protect liver and other signifiant effects.Because of the similar appearance,salers will add vegetable oil into Ganoderma lucidum spore oil soft capsule or use vegetable oil instead of Ganoderma lucidum spore oil for interests.But there are not many researches reports on quality of Ganoderma lucidum spore oil soft capsule and there are lots of adulterants can not be distinguished.So it is necessary to study on comprehensive quality researches,that could be used for evaluating quality of Ganoderma lucidum spore oil soft capsule to protect the people safe and effective medication.On the basis of previous deep study on chemical component and quality standards,we establish the comprehensive quality researches of Ganoderma lucidum spore oil soft capsule and compare it with vegetable oil.Methods1.Study on thin-layer chromatography identificationThin-layer chromatography was used to make a thin-layer qualitative identification for Ganoderma lucidum spore oil soft capsule and vegetable oil.Optimize the preparation of AgNO3-chromatoplate(the concentration of AgNO3-solution?the activation time of AgNO3-chromatoplate)and the developing the solvent system and examine the factor of temperature and humidity.2.Study on content determinationTo establish a quantitative method for determination of representative component 1.2-Dioleoyl-3-palmitoyl glycerol in the Ganoderma lucidum spore oil soft capsule and vegetable oil by HPLC-ELSD with the optimized chromatographic conditions.3.Study on the fingerprintsThe fingerprints were established of 14 batches of Ganoderma lucidum spore oil soft capsule by HPLC-ELSD,and were analyzed with fingerprint parameters common peaks and similarity analysis.Next,experimental data were analyzed with principal component analysis and cluster analysis by SPSS software.According to the common peaks and characteristic peaks,combining with the overall characteristics of spectrum,we would make distinctions between Ganoderma lucidum spore oil soft capsule and vegetable oil.ResultsThis experiment were studied and valued roundly to distinguish between Ganoderma lucidum spore oil soft capsule and vegetable oil from bothqualitative and quantitative aspects.10%AgNO3-chromatoplate was preparated by immersing method,with petroleum ether-eyhyl acetate(5:1).Results TLC spot develops clear,good resolution,reasonable Rf.The TLC spot of Ganoderma lucidum spore oil soft capsule are really different from vegetable oil's.The HPLC-ELSD chromatographic conditions were as follows:Diamonsil C18 column(250 mm×4.6 mm,5?m),mobile phase consisting of acetonitrile-isopropanol(51:49),flow rate at 1.0 mLˇmin-1.The temperature of drift tube was 47 ? the flow rate of gas was 3.2 Lˇmin-1,and the analysis time was 60 min.The average content of representative component in the Ganoderma lucidum spore oil soft capsule was 26%and in the vegetable oil(except olive oil)was between 4%to 12%.The HPLC-ELSD fingerprint of fatty acid glycerides of Ganoderma lucidum spore oil soft capsule was established.Ten peaks were confirmed as the characteristic common peaks,4 of them were identified according to the reference standards.The similarity of S1?S14 is between 0.703 to 0.998.The HPLC-ELSD fingerprint of Ganoderma lucidum spore oil soft capsule is different from the HPLC of vegetable oil.ConclusionThe comprehensive quality researches of Ganoderma lucidum spore oil soft capsule were established,the method is simple,reproducible,sensitive and specific,which will provide reference for quality control of Ganoderma lucidum spore oil soft capsule. |
