Research On Extraction Process,Quality And Initial Stability Of Ginseng In Yizhicong Granules

The Yizhicong granules,a compound preparation deriving from traditional Chinese Wendan Decoction and composed of seven herbal medicines:Ginseng,Chuanxiong Rhizoma,Acori tatarinowii Rhizoma,Aurantii Fructus immaturus,Bambusae Caulis In Taenias and etc.,has the effects of invigorating the kidney and dissipating phlegm.On the basis of retaining the original functions of regulating qi-flowing for eliminating phlegm,nourishing heart and quieting spirit,harmonizing the stomach and gallbladder,this prescription is applied to the prevention and improvement of Alzheimer's disease(AD)with significant curative effects by decomposing recipe and compatibility of medicines.Pre-pharmacological studies showed that this compound prescription highly improves the model rats' hippocampal acetylcholine transferees(ChAT)and acetylcholine(Ach)activity,repairs the NOS(Nitric Oxide Synthase)neuron injury of hippocampus of model rats,activate the cNOS,promote the synthesis of messenger molecule NO(Nitric Oxide),then increase the NGF(Nerve Growth Factor)in brain tissues of rats with Alzheimer's disease,inducing the improvement of these rats' memory.Thus,in clinical,applications of this prescription can be used for the prevention and treatment of Alzheimer's disease(AD)significantly.The main purpose of this study was to carry out pre-clinical pharmacy development research of the Yizhicong granules.L9(34)orthogonal design test was utilized to select the best percolation process of ginseng through the respective detection of the contents of Ginsenoside Rg1,Ginsenoside Re,Ginsenoside Rb1 by HPLC and the determination of the yield of dry extract,finding that the optimum percolation process was as follows:soak time was 36 hours,the concentration of ethanol was 60%,percolate volume was 10 times the amount,percolation rate was 0.5m1·min-1(equivalent to 5ml · min-1 · kg-1).Based on this prescription,the pilot experiments were conducted 3 times,the granules was analyzed of its quality,and its initial stability of ginseng in Yizhicong GranulesBased on a small scale,this research adjusted relevant parameters of production through the three-replicate pilot test to meet the needs of large-scale production.Ginseng and Acori tatarinowii Rhizoma in the Yizhicong granules were identified by TLC,and the results indicated that they could show the same color and fluorescence spots as the control or control herbs at the corresponding positions,without the interference of the negative control;Ginsenoside Rg1,Ginsenoside Re and Ginsenoside Rb1 in granules,were respectively determined of their contents by HPLC beside the methodological evaluation on the analysis method,initially establishing the quality standard of the granules and laying the foundation for the future industrialized production.
