Rhizoma Typhonii Flagelliformis is dried the tuber of Typhonium flagellifme(lodd)Blame.At present,Rhizoma Typhonii Flagelliformis is collected in the"Medicine Standard Chinese Medicinal Materials(Volume 1)of the Ministry of Health",Xin,Wen,toxic,has the effects of dampness and phlegm,used for cough and phlegm,bronchitis,etc.Sneeper,unknown swelling.Modern studies have shown that there is drug activity such as antitussive,expectorant,asthma,analgesic,anti-inflammatory,anti-allergy,inhibitory center,inhibiting saliva secretion and anti-tumor activity of pharmacological activity.Scholars at home and abroad have reported that Rhizoma Typhonii Flagelliformis contains volatile oil,amino acids,alkaloids,flavonoids and organic acids.However,at present,there are fewer research on the chemical composition and quantitative analysis of Rhizoma Typhonii Flagelliformis.In order to further clarify the basic material foundation,this article conducted the composition analysis and content measurement method of Rhizoma Typhonii Flagelliformis.The content is as follows:Research on the chemical composition of Rhizoma Typhonii Flagelliformis with chromatography.Rhizoma Typhonii Flagelliformis medicinal materials were crushed,use water decoction method to extract,use anti-phase column chromatography,large pore resin,high-efficiency liquid chromatography,and semi-preparatory liquid chromatography.The structure of the characteristics of the physical and chemical and magnetic resonance spectrum,the mass spectrum diagram and other spectrum technical appraisal compounds.As a result,17 compounds were separated from the separation of the water extraction solution of Rhizoma Typhonii Flagelliformis,namely:L-tryptophan(1),(3S)-1,2,3,4-tetrahydro-β-carboline-3-carboxylic acid(2),D-3-phenyllactic acid(3),salicylic acid(4),cyclo-(L-Ile-L-Pro)(5),dihydroconiferyl alcohol(6),cyclo-(L-Leu-L-Pro)(7),cyclo-(L-Phe-L-Pro)(8),erythro-(7R,8S)-1-(4-hydroxy-3-methoxyphenyl)-2-{4-[(E)-3-hydroxy-1-propenyl]-2-methoxyphe-noxy}-1,3-propanediol(9),isolariciresinol(10),lariciresinol(11),guaiacylglycerol-β-O-4’-synapyl ether(12),dehydrodiconiferyl alcohol-9’-O-β-D-glucopyranoside(13),dehydrodiconiferyl alcohol(14),pinoresinol(15),(+)-medioresinol(16),aurantiamide(17).Conclusion:All compounds were isolated from this plant for the first time.Using UPLC-Q-TOF-MS/MS technology combined with the UNIFI platform to qualitatively analyzes the chemical composition of Rhizoma Typhonii Flagelliformis extract.As a result,a total of 63 compounds were identified from the two modes of positive ions and negative ions,including 4 amino acid compounds,5 alkaline compounds,6 flavonoid compounds,8 phenylin and lignin compounds,6 cerebroside compounds,12 organic acid compounds,3 fatty acid glycerin compounds,2 amide compounds,2 nucleoside compounds,and 13 other types of compounds.At the same time,the cracking rules of 6 main category compounds in the six main category compounds of benzalin,phenylin,and lignin,and organis,and organic acids are summarized and summarized.The content of two compounds such as high-efficiency liquid chromatography method also measures the contents of two compounds such as dehydrodiconiferyl alcohol and pinoresinol in Rhizoma Typhonii Flagelliformis.Chromatographic conditions:YMC C18(250 mm×4.6 mm,5μm)column,mobile phase acetonitrile(A)-0.1%phosphoric acid water(B)(0~40 min,24%A),equal elution;The flow rate was 1.0 m L·min-1,the detection wavelength was 230 nm,and the column temperature was 30°C.Results:The injection amounts of the two compounds showed a good linear relationship with the peak area in the range of 0.0035~0.0878μg(R2=1.0000)and 0.0064~0.1597μg(R2=1.0000).The RSD for precision,stability,repeatability and durability is less than 2%;The mean recovery rates(n=6)were 96.19%(RSD=0.71%)and 101.03%(RSD=1.92%),respectively.This method is accurate and simple,and can be used as a quantitative detection method for dehydrodiconiferyl alcohol and pinoresinol。...
