Objective:To study the relationship between the polymorphism of CHI3L1 gene rs4950928,rs2071579,rs55700740,rs35405821,rs7515776,rs10399931 and serum YKL-40 level and the incidence of asthma in children of Guangxi Black-clothse Zhuang,and the distribution frequency characteristics of polymorphisms of the above 6 loci in children with Guangxi Black-clothse Zhuang asthma,and the results were Compared with children of other races and han nationality.To investigate the relationship between the polymorphism of CHI3L1 gene and the susceptibility of children with Guangxi Black-clothse Zhuang asthma.Methods:With the help of SnaPshot SNP typing method,CHI3L1 gene polymorphisms at rs4950928,rs2071579,rs55700740,rs35405821,rs7515776 and rs10399931 of 240 subjects(60 Black-clothse Zhuang healthy children,60 Black-clothse Zhuang asthmatic children,60 healthy children of the han nationality and 60 asthmatic children of the han nationality)were genotyped.Meanwhile,enzyme-linked immunosorben assay(ELISA)was used to detect the serum YKL-40 level.SPSS17.0 statistical software was used for data statistical analysis.The genotype and allele frequency of the asthma group and healthy control group of children with Black-clothse Zhuang nationality,and the asthma group and healthy control group of children with han nationality were calculated by counting method,and Hardy-Weinberg law of genetic balance was used to test population representativeness.The genotype frequency and allele frequency of chi-square test compared groups.Serum YKL 40-level with x±s,serum level of comparison between different groups using independent sample t test,all the above statistical analysis,P < 0.05,the difference was statistically significant.Results:(1)In the above six loci of CHI3L1 gene in the Black-clothse Zhuang children,except rs4950928 loci only CG and CC genotypes were detected in the han asthma group,three different genotypes were detected in the other five loci in the four groups of subjects.Among them,CG,CC and GG genotypes were detected at rs2071579 and rs35405821,GT,GG and TT genotypes were detected at rs55700740,AA,AT and TT genotypes were detected at rs7515776,and CT,CC and TT genotypes were detected at rs10399931.(2)There were racial differences in allele frequency distribution of the above 6 loci.(3)The serum YKL-40 level in the asthma group was significantly higher than that in the healthy group,and the difference was statistically significant.the serum YKL-40 level in the han asthma group was higher than that in the Black-clothse Zhuang asthma group,and the difference was statistically significant.The above results are different from the relevant literature reports,which may be related to race,ethnicity,geographical difference and limited number of samples,this study reflects the relationship between CHI3L1 gene and Black-clothse Zhuang asthma,which further enriches the gene pool.Conclusion:(1)Polymorphisms exist in CHI3L1 gene loci rs4950928,rs2071579,rs55700740,rs35405821,rs7515776 and rs10399931 in Black-clothse Zhuang children,which may not be related to asthma susceptibility in Black-clothse Zhuang children.(2)The allele frequency distribution of CHI3L1 gene loci rs4950928,rs2071579,rs55700740,rs35405821,rs7515776 and rs10399931 in Black-clothse Zhuang children showed racial difference,but the polymorphism,genotype frequency and allele frequency distribution showed no national difference compared with that of Chinese han children.(3)Elevated serum YKL-40 level may be related to the incidence of asthma in Black-clothse Zhuang children.And the serum level of YKL-40 in asthmatic children of han nationality showed national difference.
