Objective:to establish a C57BL/6 mouse model of H.pylori infection related gastritis by gavage of NaHCO3 and h.pyloris1(Helicobacter pylori Sydney strain 1)strains.The pathological changes of gastric mucosa,the clearance of H.pylori,the detection of ELISA,immunohistochemistry and Western blot were studied The changes of inflammatory factors in mice before and after the treatment of different groups of drugs were fully observed,the therapeutic effect of xinpuyin combined with omeprazole on H.pylori-related gastritis was objectively evaluated,and a new method of treating H.pylori-related gastritis with traditional Chinese medicine as the main and Western medicine as the auxiliary was explored.Methods:H.pyloris 1 strain(standard strain of mouse infection)was inoculated into the culture medium containing defibrillated amniotic blood under appropriate mixed gas,and the culture lasted for 2-3 days,during which the bacterial culture was observed continuously.After the culture was identified as H.pylori positive,the bacterial solution was made for standby.Fifty C57BL/6(B6)mice were randomly divided into control group,model group,xinpuyin group,xinpuyin combined with omeprazole group,amoxicillin+kelamycin+omeprazole enteric coated tablet group(gastric triad group)and control group,After that,each group was given 14 days continuously,the control group and the model group were given the same dose of saline.Four weeks later,the mice were killed.According to the experimental requirements,the corresponding tissues and serum were taken out for preservation.The distribution and colonization of H.pylori were observed by rapid urease method and Giemsa staining method.The expression of IL-8,immunohistochemistry and Western were detected by ELISA The protein expression of IL-8,NF KB and other inflammatory factors was detected by blot,and the mRNA expression of IL-8,NF KB was detected by PCR.Results:compared with the control group,H.pylori infection was found in the gastric mucosa of the model group;compared with the model group,H.pylori infection in the gastric mucosa of the mice in each administration group was significantly reduced,and the gastric triad group was more obvious than the Xin'ao group,followed by the Xin Puyin group;before treatment,compared with the control group,the related inflammation in the model group was found After treatment,the protein,mRNA and serum levels of IL-8 and NF kb in xinpuyin group,Xinao group and weisanlian group were significantly lower than those in the model group(P<0.05)?Conclusion:animal experiments show that xinpuyin can treat H.pylori-related gastritis,and the combination of omeprazole is more effective,which is similar to the triple effect of stomach.Therefore,xinpuyin combined with omeprazole is expected to be a new method for the treatment of H.pylori-related gastritis.
