Background and purposeAsthma,originally from the medical master zhu danxi's book "treatment of symptoms and pulse",in the synopsis of the golden chamber written by zhang zhongjing of the han dynasty,it is called "shangqi" and described as follows: "cough and shangqi,water chicken in the throat",it's similar to the wheezing that we hear in modern medicine.Modern medicine believes that asthma is caused by a variety of active inflammatory mediators,including molecules,cells and cytokines,which are used in the body to cause respiratory tract diseases with high response;chronic airway mucosal inflammation,high responsiveness,irreversible damage as the main characteristics,endangering the health of different age groups,different races and even life around the world,has become a serious public health problem.Now the number of children with asthma worldwide are increased significantly,the pathogenesis of the disease is not fully clear,however,and many of the children and their parents do not have a good understanding of the hazards and causes of asthma,lead to many children not according to the correct standard medication,not systematic and standardized treatment,lead to asthma attack repeatedly,that irreversible damage occurs,psychological,physiological and growth and development of children have a great impact,hence,at present,how to timely control and effectively prevent asthma attacks and assess the severity of children's disease more easily,become the urgent problems in the clinical work.It is known that the pathologic progression of asthma is characterized by prolonged airway mucosal inflammation,high responsiveness,and irreversible damage,is essentially a variety of inflammatory factors stimulate the airway to produce corresponding pathological changes,leading to different degree of asthma attacks,therefore,it is of certain significance to detect the changes of related factors in the process of airway diseases.Hypoxia inducing factor 1(HIF-1)is produced by a lack of oxygen environment,belong to dependent on the oxygen activation of transcription factors,by the beta of structural expression and expression under the condition of alpha subunit composition in the form of dimers,and HIF-1 alpha is the main function of the unit.In normal body environment,have very low levels of HIF-1 alpha,in the body hypoxia environment,HIF-1 alpha activation,its expression level increased significantly,through to the role of the target genes to make the body produces a series of compensatory reaction,involving many aspects,including the formation of new blood vessels,the number of red blood cells increased,cell proliferation fibrosis,regulate the diameter of blood vessels to improve the ability to adapt to hypoxic conditions;and vascular endothelial growth factor(VEGF)is one of the active site factors of HIF-1alpha,is now the cognitive scope with the generated form new blood vessels in the main stimulating factor,the effect on receptors of alveolar epithelial cell,airway smooth muscle cells and vascular endothelial cells,lead to endothelial vascular permeability increase,vascular intimal thickening of the blood vessel,stenosis,generate new blood vessels;so HIF-1 alpha and VEGF are both involved in the pathological processes of airway mucosal inflammation,high response and remodeling.As a result,we consider that the changes of HIF-1alpha and VEGF have important implications for the severity of childhood asthma.Objective: explore hypoxia inducing factor 1 alpha(HIF-1?),vascular endothelial growth factor(VEGF)on the pathogenesis of childhood asthma process function and changes in serum,study the correlation of the two factors and the degree of the cases.MethodThe children in the asthma group were hospitalized in the pediatric ward of our hospital from November 2018 to October 2019.According to the results of lung function and the severity of the disease measured,the children in the asthma group were further divided into the mild children group and the moderate and severe children group;Control group subjects,are from the same period in the center of the medical health physical examination of children,Serum levels of HIF-1alpha and VEGF were detected by ELASA method,HIF-1alpha levels and VEGF expression levels were compared between the two groups and the two subgroupscontrast,then asthma group of subjects is analyzed on the son of two factor level change and its possible relationship between disease severity.Result1.The expression level of serum HIF-1 alpha and VEGF in children with asthma was significantly higher than that of normal subjects,and the difference was statistically significant(P<0.001).2.The expression levels of HIF-1 alpha and VEGF in the mild and moderate children subgroup were all higher than those in the normal children group,and the HIF-1 alpha and VEGF expression levels in the moderate and severe children subgroup were more significant,that is,the more severe the disease,the higher the HIF-1 alpha and VEGF expression levels were,and the difference was statistically significant.(P<0.001).Conclusion1.The expression level of HIF-1 alpha,VEGF in the serum of asthmatic children was significantly higher than that of healthy children,which was closely related to the incidence of asthma.2.The serum expression levels of HIF-1 alpha and VEGF in asthmatic children were positively correlted with the severity of the disease,and that is,the higher the expression level of the two factors,the more severe the disease.
