This thesis mainly includes two aspects of researches.The first one investigates the systematic phytochemistry and bioactivity of Lilium lancifolium Thunb flowers.While,the second one studies the chemical composition and hypoglycemic activity of the n-butanol from Patrinia scabiosaefolia extracts.First of all,an overview about the latest research progress on the chemical constituents of Lilium from 1989 to now was discussed in a review.It was conducive to a systematic study on the chemical components and bioactivities of Lilium.In addition,the chemical constituents of the flowers of L.lancifolium were isolated by different types of chromatography: silica gel column chromatography;gel column chromatography and preparative chromatographic column.The main compounds were identified as Kaempferol(1),Quercetin(2),Tamarixetin 3-O-?-D-glucoside 7-O-?-L-rhamnoside(3),?-sitosterol(4),Isoquercitrin(5),Caffeic acid(6),Daucosterol(7),Quercetin-3-O-?-D-galactopyranside(8).Among them,the compound Tamarixetin 3-O-?-D-glucoside 7-O-?-L-rhamnoside was isolated from the plant for the first time.The effect of Kaempferol,caffeic acid and Quercetin-3-O-?-D-galactopyranside on glucose absorption in Caco-2 cells were investigated.Glucose determination kit and 2-NBDG were used to detect Caco-2 cells glucose uptake and consumption.Glucose determination kit results showed that caffeic acid and quercetin-3-O-?-D-galactopyranside could promote the glucose consumption in normal Caco-2 cells.The 2-NBDG results showed that caffeic acid and quercetin-3-O-?-D-galactopyranside promoted the glucose absorption in normal Caco-2 cells.Western blot was used to detect SGLT1 and GLUT2 related proteins expression changes.The results indicated that the caffeic acid and quercetin-3-O-?-D-galactopyranside can increase normal cells SGLT1 and GLUT2 protein expression level at high doses(200 ?M).Caffeic acid and quercetin-3-O-?-D-galactopyranside may regulate glucose transport by mediated the expression of SGLT1 and GLUT2 and promote glucose absorption in Caco-2 cells.Secondly,regarding to previous researches,the ethyl acetate fraction of P.scabiosaefolia was isolated and a large number of iridoid glycosides was identified.In this study,variety of chromatographic materials and extensive spectroscopic methods(NMR,UV,IR,HSQC,HMBC,1H-1H COSY,ROESY)were used to isolate and identify the constituents in the n-butanol fraction of P.scabiosaefolia.A total of ten compounds were isolated and identified as follows: ?-sitosterol(9),daucosterol(10),ursolic acid(11),3?-hydroxy-12-ursene(12),patriscabioin G(13),blumenol A(14),eudesmin(15),sweroside(16),patriscabioin A(17),patriscabioin(18).Moreover,their ability to hypoglycemic activity was assessed by the method of MTT and glucose oxidase-peroxidase.Results showed that patriscabioin A(17)could significantly improve glucose consumption in insulin-resistant 3T3-L1 adipocytes at the concentrations of 25 ?M and 50 ?M(P < 0.001)compared with the model group.Compared with the compound 17,patriscabioin(18)could significantly decreased the glucose content in insulin-resistant adipocytes at the concentrations of 12.5 ?M,25 ?M and 50 ?M(P < 0.001).
