Exercise fatigue refers to the physiological process of the body can not maintain the predeterminded exerciseintens ity.Astaxanthin is a natural antioxioxidant.Studies have shown that astaxanthin has anti-fatigue effect,metabolomics method can be used to study astaxanthin from the level of small molecule metabolites anti-fatigue mechanism of action.Objective: Use the metabonomics method to explore the effections on the metabolic characteristics of adding natural astaxanthin to the rat before and after have an exhaustive exercise.Methods: 48 male rats were randomly divided into control group(C),medicine group(M),exercise group(E),take medicin immediately after exercise(EMJ),take medicin 12 h later after exercise(EM12),take medicin 24 h later after exercise(EM24).The rats in the medicine group were fed with astaxanthin continuously for 21 days on a day of 6.6mg/kg,and the exercise rats were loaded with swimming exercise to fatigue for three weeks.The rats' blood of EMJ,EM12,EM24 were got from the femoral artery and centrifugal preservation,then the NMR data of blood samples were collected and processed,then it will be analyzed by Mest Re Nova software,SIMCA-P13.0 software and SPSS17.0 software.At last,combine the P value and VIP value to find out the difference of metabolism.Results:(1)The density in rats' serum for M group of acetoacetate,glutamic,acid,creatine(P < 0.05),N-acetyl-glycoprotein,leucine,isoleucine,3-Dhydroxy butyric acid(P < 0.01)increased significantly compared with C group,trimethylamine oxide(P < 0.05),lactic acid,glycerol(P < 0.01)were significantly decreased.(2)The density in rats' serum for E group of glycerol,trimethylamine oxide(P < 0.05),lactic acid,acetoacetate(P < 0.01)increased significantly compared with C group,glucose,lipid,leucine(P < 0.05),isoleucine,valine,alanine,creatine,betaine(P < 0.01)were significantly decreased.(3)The density in rats' serum for EMJ group of serum glucose(P < 0.05),leucine,isoleucine and valine(P < 0.01)increased significantly compared with group E,while lipid(P < 0.05),lactate and glycerin(P < 0.01)decreased significantly.(4)The density in rats' serum for EM12 group of serum leucine(P < 0.05),glucose,isoleucine,valine and betaine(P < 0.01)increased significantly compared with group EMJ,and the concentration of glycerol(P < 0.05),lactate and trimethylamine(P < 0.01)decreased significantly.Conclusions:(1)The supplement of astaxanthin can increase the level of glucose in the body,promote the fat decomposition and increase the content of BCAA.(2)Fatigue exercise can accelerate the decomposition of sugar,fat and protein,and accumulate the lactic acid.The supplement of astaxanthin can maintain the blood glucose level of the body,reduce the accumulation of lactic acid,promote the energy supply of lipid,save the use of BCAA and postpone the fatigue state.(3)The rats with astaxanthin after fatigue exercise 12 h will weaken the demand for glucose,fat and amino acid metabolism compared with the rats have fatigue exercise immediately.
