Objective: This study selected Rhodococcus erythropolis(R.erythropolis)as a model,studied whether the gut microbiota contain LDs,isolated and proteomic analyzed of LDs from R.erythropolis to determined the major protein of LDs,then used this protein to found possible LD-containing gut microbiota and analyzed correlation between possible LDcontaining gut microbiota changes with different exercise intensities of female swimmers.The study may provide information for improving the athletes' body health and sports performance.Methods: In this experiment,R.erythropolis growth curve was detected and established.The relative contents of triacylglycerol(TAG)and proteins were determined by enzymatic analysis,and then the components of neutral lipids were analyzed by thin layer chromatography(TLC).The neutral lipids then stained and their stainingt structures were observed by confocal microscopy.These three methods are used to determine possible lipid droplets in R.erythropolis.Then LDs were isolated from R.erythropolis and subjected to lipidomics and proteomics.The main proteins were fused by green fluorescent protein(GFP)and imaged by confocal microscopy.The bioinformatics methods were utilized to discover more bacteria owning LDs.The data of 16 S rRNA high-throughput sequencing from the gut microbiota database of female swimmers under different exercise intensity were finally analyzed.Results: 1)The intestinal bacteria R.erythropolis grew to saturation in 36 hours.2)TAG accumulation from R.erythropolis reached to top at 36 hours.3)TAG was the major neutral lipid in R.erythropolis and their LDs.They also had other neutral lipids and phospholipids.4)Two samples were subjected for shotgun analysis,and the results of proteomics analysis were obtained.A total of 224 LD proteins was found and then divided into eight groups according to their functions.5)PspA was located on LDs.6)All 33 actinomycetes may contain lipid droplets.7)The relative abundance of possible LDcontained gut microbiota was higher in high-intensity than in low-intensity athletes.Conclusion: 1)The intestinal bacteria R.erythropolis has LDs.2)PspA was located on LDs.3)All 33 gut microbiota collected may contain LDs.4)The relative abundance of possible LD-contained gut microbiota was higher in high-intensity than in low-intensity athletes.
