Objective: To investigate the effects of Xiaoshuanyin on TM and m EPCR on femoral vein endothelial cells in traumatic deep vein rabbits.Methods: Thirty-six healthy adult male New Zealand white rabbits were randomly numbered into four groups according to their weighted stratification,which were A(Xuanshuan group),B(low molecular heparin sodium group),C(normal saline group),and D(Sham operation group),9 in each group;of which three groups A,B,and C use the clamp and plaster external fixation to reproduce the rabbit femoral vein thrombosis model;group D made the same surgical incision as the first three groups;Drug treatment,4 hours after the second intragastric administration on days 1,3,and 7 after surgery,3 femoral vein blood vessels were randomly collected from each group and stored in-80 ° C.After thawing,grinding,After centrifugation,the Elisa method was used to detect the changes of TM and m EPCR content on the femoral vein vascular endothelial cell membrane of rabbits in each group.The data were collected and statistically analyzed.Results: During the entire experiment,there was no significant difference in the TM content level and m EPCR content level in the sham operation group in group D at all stages(PD>0.05),indicating that the sham operation group did not form thrombus,the vascular endothelial cells were intact,and the barrier function was stable.There was no significant change in the data of the two indicators during the process.The contents of TM,m EPCR in each of the three groups A,B,and C were lower than those in group D.Among them,the contents of TM and m EPCR in group A were lower than those in the low-molecular-weight heparin sodium group in the first day after surgery(?P<0.05).There was no significant difference in the normal saline group in group C(?P>0.05),indicating that low-molecular-weight heparin sodium had a faster effect than Xiaoshuan Yin at the beginning of the experiment,and there was no significant difference between the two groups on days 3 and 7 after surgery(?P>0.05),which is much higher than TM in group C,m EPCR expression(?P <0.05).It shows that Xiaoshuanyin and low molecular weight heparin sodium can promote the expression of TM and m EPCR on vascular endothelial cells.The TM and m EPCR of group C showed a slowly rising state during the experiment.The comparison between 1d and 3d and 7d after operation showed significant differences in data(Pc1<0.05,Pc3 <0.05);the comparison between 3d and 7d after operation was intuitive There is a slight increase on the 7th day,but there is no significant difference(PC2>0.05);it may be the effect of autoanticoagulation.Vascular endothelial cells will try to maintain their own barrier function by promoting TM expression,but this function is on the 3rd to the 3rd day.7d may reach its peak.Conclusion: 1.Through rabbit femoral vein forceps combined with gypsum external fixation,a DVT rabbit model was successfully manufactured.2.In the early stage of vascular endothelial cell injury,the levels of TM and m EPCR on the cell membrane will be reduced by the body's own coagulation reaction,and the influence of inflammatory factors will be reduced.3.In the protein C system-mediated anticoagulation pathway,Xiaoshuanyin can promote TM and m EPCR levels on vascular endothelial cell membranes to promote the activation of protein PC,thereby inactivating coagulation factors ?and ? to exert anticoagulation The effect of Xiaoshuanyin is slower than that of low-molecular-weight heparin sodium in the whole treatment process,but in the end there is no significant difference between the two effects.
