Acute high altitude pulmonary edema(HAPE)has rapid onset,rapid development,and severe symptoms.At present,the therapy is inconvenient and has many side effects.It has become a common disease that seriously endangers the health and life of people who go to the high altitude.Therefore,increasing research on HAPE and enriching treatment methods is of great significance for reducing the incidence of disease and slowing the disease process.Plaster acupuncture therapy is an innovative therapy developed by using high-quality Sibin vermiculite as raw material and using modern advanced ultra-fine powder coating technology.It has the advantages of strong timeliness,wide treatment range,portability,greenness,safety,and no side effects,which greatly expands its application breadth.In this study,plaster acupuncture therapy was used to stimulate the "Baihui" and "Neiguan" points in rats.The effects of this therapy on oxygen free radicals,inflammatory factors,and lung water content in rats were observed.ObjectiveIn this experiment,rats with acute high altitude pulmonary edema model established under low pressure and hypoxia for 24 hours were used as research objects.Dazhu Hongjingtian Capsule was used as the positive control group.The "Neiguan" and "Baihui" acupoints were used as treatment points.Pre-treatment with plaster acupuncture for 14 days before entering the cabin.To observe the effects of plaster acupuncture therapy on SOD,MDA,CAT,GSH-PX,IL-6,TNF-?,lung water content and lung pathological morphology in rat serum and lung tissue.To explore the preventive and therapeutic effects of plaster acupuncture therapy on high altitude pulmonary edema model rats,the effects of plaster acupuncture therapy on lung tissue and serum free radicals and serum inflammatory factors in low pressure and hypoxia model rats,and to explore the effect of PLASTER ACUPUNCTURE therapy on prevention and treatment of plateau in model rats Possible mechanism of pulmonary edema.MethodForty-five male SD rats were randomly divided into normal group,normal group+plaster acupuncture,model group,model group+plaster acupuncture,model group+drug,9 rats in each group.At "Neiguan" and "Baihui",the model group was affixed with adhesive tape for 1 h every day,the model group+plaster acupuncture and the normal group+plaster acupuncture were affixed with adhesive tape for 1 h every day and fixed with adhesive tape.Dazhu Hongjingtian was administered at a dose of 280 mg/kg once a day,and the pretreatment in the four groups lasted for 14 days.After the pretreatment was completed,except for the normal group and the normal group and the normal temperature and pressure environment,the other three groups were placed in a simulated plateau low-pressure oxygen cabin on the 15th day for 24 hours.Blood was taken from the heart after exiting the chamber.The lung tissue and serum SOD,GSH-PX,CAT activity and MDA content were measured by colorimetric method.The levels of IL-6 and TNF-? in serum were measured by enzyme-linked immunosorbent assay.Water volume and gross and pathological sections of the lung were observed.Results1.Comparison of general conditions:After leaving the cabin,the model group responds slowly,while the model group+plaster acupuncture therapy,model group+drug performance is acceptable.2.During the pretreatment process,the weight of the rats in each group increased steadily.Statistical analysis of the growth of each group showed that there was no significant difference(P>0.05),indicating that pretreatment had little effect on the weight gain of each group.After 24 hours of fasting,the normal group lost less weight on the 14th day,and the rest of the other groups placed in the simulated high-pressure hypobaric oxygen cabin on the 14th day showed significant weight loss.Compared with the model group,the model group+plaster acupuncture and the model group+drug weight decreased less(P<0.05),and there was no significant difference between the two groups(P>0.05).3.Rat serum SOD,GSH-PX,CAT and MDA:Compared with the normal group,SOD,GSH-PX,and CAT in the model group were reduced(P<0.05)and MDA was increased(P<0.05).Compared with the model group,SOD,GSH in the model group+plaster acupuncture therapy rats SOD?GSH-PX,CAT significantly increased(P<0.05),MDA content decreased significantly(P<0.05).There was no significant difference between the model group and the patch and the model group and the drug(P>0.05).4.SOD,GSH-PX,CAT and MDA in rat lung tissues:Compared with the normal group,SOD,GSH-PX,and CAT in the model group were all reduced(P<0.05),and MDA was increased(P<0.05).Compared with the model group,SOD,GSH-PX,and CAT in the model group+plaster acupuncture were significantly increased(P<0.05),and the MDA content was significantly reduced(P<0.05).There was no difference between the model group+plaster acupuncture and the model group+drugs.Significant difference(P>0.05).5.IL-6 and TNF-? in rat serum:Compared with the normal group,serum IL-6 and TNF-? in the model group were increased(P<0.05).The serum levels of IL-6 and TNF-? in plaster acupuncture rats were significantly reduced(P<0.05);there was no significant difference between model group+ plaster acupuncture and model group+drugs(P>0.05).6.Water content of lung tissue in rats:Compared with the normal group,the water content of lung tissue in the model group increased(P<0.05).Compared with the model group,the serum lung water content in the rats of the model group and the patch was significantly reduced(P<0.05).There was no significant difference between the model group and the patch and the model group and the drug(P>0.05).7.General condition of the lungs:The lungs of the normal group,the normal group and the patch rats had smooth lung surface,pale red color,and good elasticity.In the model group,the lungs were full and swollen,the lung membrane was tense,the color was dark red,and there was a little piece of stasis and bleeding under the capsule.The model group+plaster acupuncture had smooth lung surface,pale red color,good elasticity,and few bleeding points in the capsule;In the model group,the lungs were slightly swollen,reddish,and there were scattered bleeding points under the capsule.8.HE staining section:normal congestion,edema and inflammation were not observed in normal group and normal group+plaster acupuncture lung tissue structure;In the model group,there were a large number of inflammatory cells infiltrating the lung interstitial and alveolar cavities,vascular congestion in the lung tissue,and tissue edema.Model group+plaster acupuncture showed mild tissue edema and inflammatory cell infiltration.Model group+drugs showed mild tissue edema and inflammatory cell infiltration,alveolar epithelium hyperplasia,alveolar wall thickening,widened interval,lighter edema,and local tissue cell proliferation.Compared with the normal group,the alveolar septum in the lung tissue of the model group was widened and the pulmonary interstitial edema.The model group+plaster acupuncture effectively reduced the alveolar interstitial widening and pulmonary interstitial edema caused by hypoxia.Conclusion1.1 Plaster acupuncture therapy has a certain protective effect on the lungs of rats with hypotension and hypoxia.First,plaster acupuncture therapy can increase the speed and magnitude of weight gain in model rats.Second,plaster acupuncture therapy can reduce the lung tissue water content in model rats,that is,reduce the lung tissue edema.Third,it can be seen from pathological sections that plaster acupuncture therapy can improve the pathological morphology of model rats' lungs,reduce the alveolar interstitial widening,inflammatory cell infiltration,alveolar wall thickening,pulmonary interstitial edema,inflammatory cell infiltration,local Histiocytosis and alveolar epithelium.1.2 Plaster acupuncture therapy can effectively increase the vitality of SOD,GSH-PX,and CAT in lung tissue and serum,and reduce the content of MDA,suggesting that the patch generally improves the ability to scavenge oxygen free radicals and adjusts the oxidative stress state of lung tissue and the body.1.3 Plaster acupuncture pretreatment can effectively reduce the serum levels of IL-6 and TNF-a in model rats,reduce the body's inflammatory response,and control further lung injury in acute hypobaric hypoxia model rats.1.4 Plaster acupuncture pretreatment can effectively reduce the damage to lung tissue,thereby reducing pulmonary edema,pulmonary membrane tension,and subcapsular hemorrhage.1.5 Plaster acupuncture therapy has a certain prevention and treatment effect on the body injury caused by acute low pressure and hypoxia,and the treatment effect on model rats is not significantly different from that of Dazhu Hongjingtian capsule.Its mechanism of action may be to stimulate the "Baihui" and "Neiguan" acupoints through plaster acupuncture therapy,reduce the expression of oxygen free radicals and inflammatory factors,and adjust the oxidative stress state of the lungs and the whole body to promote lung tissue and serum.The expression of MDA is reduced and the levels of serum IL-6 and TNF-? are reduced,lung tissue edema is reduced,the increase of blood oxygen supply and the reduction of interstitial fibrosis form a virtuous circle,and finally alleviate the damage caused by acute hypotension and hypoxia To improve the symptoms of acute high altitude pulmonary edema in patients.
