Object:Chronic fatigue syndrome(CFS)is a severe sub-healthy condition characterized by long-term persistent fatigue,accompanied by poor concentration,memory loss,fever,headache,sore throat,and muscle and joint pain with a group of non-specific symptoms such as sleep disorders and depression.CFS is more common among middle school students,affecting their physical health and learning.Due to the current lack of understanding of the physiological mechanisms of CFS,it is difficult to explore the rehabilitation methods of CFS.In this study,a non-invasive detection method was used to confirm the health status of the body:detection of 3-nitrotyrosine(3-NT)content changes and nail keratin functional group variation peaks confirmed its high protein oxidation status.The use of the First Beat at Bodyguard 2 to monitor heart rate variability(HRV)characteristics,stress and fatigue recovery 24 hours after exercise and 24 hours after exercise intervention confirmed the disorder of autonomic nerves,high learning pressure,and poor recovery from fatigue.Based on the above-mentioned characteristic information,the physiological mechanism of CFS is explored,and the characteristics of physiological changes on the students of CFS are explored to provide a theoretical basis for in-depth study of CFS.Methods:According to the CDC-94 CFS criteria from the U.S.Centers for Disease Control and Prevention,20 CFS patients were selected from 15 schools in Guanzhong area,and 20 healthy middle school students were randomly selected as the normal control group.?Using the "Physical Health Test Method" to test the lung capacity,1000 meters running,grip strength,standing long jump,pull-ups and seated body flexion levels of the two groups of students.? The nails of the CFS and control groups were collected and washed sequentially with 0.15 mol/L sodium chloride(24 hours),water,and ethanol and dried before being tested.3-nitrotyrosine content test:Take 40?g of washed sample,rinse with 5mL 2%sodium dodecyl sulfate(SDS)and 50mmol/L sodium phosphate(pH 7.8),and after drying,Re-immersed in a mixture of 5 mL of 2%SDS,50 mmol/L sodium phosphate(pH 7.8)and 20 mmol/L dithioerythritol(DTE)and incubated at 65? overnight.The sample was stirred for 1 h at room temperature with a magnetic stirrer,centrifuged(15000 g)for 20 min,and the supernatant was taken.The OD of the supernatant was measured using an ultraviolet-visible spectrophotometer,and the concentration of the sample was calculated indirectly.Keratin functional group variation peak detection:Take 3-5 mg of cleaned sample and mix it with 400 mg of potassium bromide,grind it in an agate mortar for 5 to 10 minutes,compress the tablet,and then select 500 to 4000 cm-in the infrared spectrometer.The tested data was imported into Origin 8.5 software for analysis.? Connect Bodyguard's electrodes and equipment,and place the device on the student's body.The instrument's large electrode is placed under the body's right clavicle and the small electrode is placed under the body's left rib.After 24h,remove the device,upload data through Firstbeat compatible software,connect the device to the computer's USB port and open compatible software to start reading and exporting data.? This experiment uses a modified step test and Wingate's anaerobic power combination as exercise intervention.Before the exercise,the urine indexes of the two groups of students were collected;during the exercise,relevant indicators for evaluating the exercise ability were collected;after the exercise,the urine indexes were collected and the body condition monitoring was performed for 24 hours.The physical status monitoring method 24h after exercise intervention is the same as above.Results:? Results of physical fitness tests showed that the results of the 1000-meter run in the CFS group were 247.70 4.35 s,while those in the control group were 226.50 ±6.20 s.There was a significant difference between the two groups.There were no significant differences in the results of the other five test items.? The 3-NT content of nails in the CFS group was 47.74±1.76 pmol/g,and that of the control group was 38.42±1.75 pmol/g.There was a significant difference between the two groups.The 3-NT content in nails of CFS middle school students was significantly higher than that of normal middle school students.The keratin infrared spectra of CFS students showed significant differences at the 4 absorption peaks:the peak intensity at 3556.1 cm-1 and 1185.6 cm-1 weakened;the absorption peak at 3232.6 cm-1 and 2815.3 cm-1 disappeared.? The RMSSD of the adjacent RR intervals of CFS middle school students was 38.69±71.87 ms,the standard deviation of RR interval(SDNN)was 126.50±5.10 ms,and the low frequency power(LF)was ms2.The power(HF)was 779.00±24.29ms2,and the LF/HF was 2.04±0.04.In the control group,the RMSSD was 44.53±1.91ms,the SDNN was 141.50±2.90ms,the LF was 1456.00±20.35ms2,and the HF was 927.50±21.84 ms2.LF/HF is 1.59±0.03.The average sleep time of students in the CFS group was 6.89±0.20 h,and the fatigue recovery index was 107.20±3.48.The average sleep time in the control group was 7.40±0.18 h,and the fatigue recovery index was 123.10±4.28 h.The HRV,nighttime sleep,and fatigue recovery index were significantly different between the two groups of students.The HRV,sleep duration,and recovery index of students in the CFS group were lower than those in the normal group.??After 24 hours of exercise intervention,the RMSSD of the students with chronic fatigue syndrome was 31.15±1.39ms,the SDNN was 118.50±5.01ms,the LF was 1573.00±53.55ms2,the HF was 806.50±38.74 ms2,and the LF/HF was 1.99±0.08;the control students The RMSSD was 42.00±1.60ms,the SDNN was 150.90±3.82ms,the LF was 1457.00±24.82ms2,the HF was 893.90±18.38ms2,the LF/HF was 1.61±0.02;the CFS student's fatigue recovery index was 97.20±2.38,the control group The recovery index is 104.40±2.41.The HRV and recovery index of the two groups of students had extremely significant differences.The CFS students had lower HRV and recovery index than the normal group.Conclusion:?The results of the physical fitness test show that the CFS students have poor physical health,aerobic endurance is poorer than the control group especially.?The results of the level of body oxidation show that compared with the control group,the degree of oxidation in the body is higher and there are different degrees of damage in the chemical bond of the protein in the CFS students' body.?The CFS students have high heart rate,low heart rate variability,less sleeping time,poor sleep quality,and small fatigue recovery index in daily life,which indicate that their daily stress is high,however,their relaxation time is not enough.Their autonomic nervous system may be disordered.?CFS Students' aerobic exercise ability and anaerobic exercise ability are worse than those in the control group.The CFS students have high heart rate,low heart rate variability,low fatigue recovery index,and improved sleep time after 24 hours of exercise intervention,which indicate that this kind of exercise is intense for students of CFS.The CFS student's body condition cannot return to normal within 24 hours.It also shows that exercise can promote sleep but cannot significantly improve sleep quality.
