The sera of 392 individuals have been investigated for the presence of the Down Syndrome Protein (DSP) by a modified immunoelectrophoretic procedure. The results showed that 64.2% of the couples with DS affected children were DSP positive which was well in accordance with the reported occurrence of nondisjunction at meiosis I from various research groups (53.8% to 90.3%). Furthermore, from the study of 56 pure meiotic nondisjunction trisomy cases, 40 cases (71.4%) showed that the DSP was present in the sera of the parents who were responsible for the extra chromosome in the DS affected children. This evidence strongly supported the original hypothesis by Miles that the presence of the DSP was associated with nondisjunction occurring during the first meiotic division.;From lipid profiles of 36 participants, a significant increase in concentration of serum total cholesterol was found in the DSP positive donors (at P ;The DSP has been proven to be a variant of apolipoprotein B with an increased electrophoretic mobility. The author proposes that the increased electrophoretic mobility of the DSP may be due to LDL peroxidation in vivo. The results of a heat study showed that heat caused the apolipoprotein B to mimic the DSP pattern, which suggested that lipid peroxidation may have been initiated in the serum after heat incubation, or in vivo antioxidant systems failed to function properly.;The concentrations of two serum antioxidants, transferrin and ceruloplasmin, were measured by rocket electrophoresis.
