Medicinal tea is a single product or a combination product,which is processed from plant leaves(buds),flowers(buds),rhizomes,fruits and other raw materials used in medicine and food.They are served by tea brewing and boiling.As a new type of herbal tea,Z.jujuba leaves play a important role in antioxidant,soothe the nerves and promote sleep.Compared with traditional tea,it does not contain caffeine,theophylline and other irritating or refreshing compounds,and has become a hot research material in the current academic circle.In this study,we takes the Z.jujuba leaves from the cultivation base of Tianjiaohong Agricultural Technology Co.,Ltd.as the research object.Qualitative analysis and quantitative calculation of 5 chemical components in 5 kinds of Z.jujuba leaves samples in different collection periods by HPLC method.Use SPSS to perform One-Way ANOVA Tukey significance comparison and ROC analysis on the data obtained from the HPLC experiment.To explore the change law of the main chemical components in Z.jujuba leaves and determine the varieties of Z.jujuba leaves for medicinal tea and the best picking time.At the same time,it provides important research foundation and data information for the production of medicinal tea.Research indicates:(1)Determined the HPLC analysis method.Gradient elution of 0.2%phosphoric acid-0.2% triethylamine aqueous solution,water(A)and acetonitrile(B).Column temperature 30?,flow rate 1m L/min,injection volume 40 ?L.Establish an elution system with different chemical components.Rutin,Hyperoside,and Isoquercitrin using the same elution method.Protocatechin,Catechin using another elution method.(2)Determined the optimal extraction conditions.To verify its effectiveness by carrying out repeated experiments of methodology.Experimental data shows: All five compounds have high linear correlation coefficients(>0.999)when tested.The RSD values of the intra-day and inter-day precision of the test were both <1.00%,and the distribution was normal.Repeatability and stability in the range of 120 h,the RSD value is only 1.59%.The recovery rate of adding reference substance is higher than97%.The maximum RSD is only 1.16.This method performs well in experiments such as sensitivity,accuracy,and repeatability.(3)According to the results of statistical analysis: The main ingredient in Z.jujuba leaves is flavonoids(Rutin),the content range is249mg/100g-80mg/100 g.The content of Protocatechin and Catechins is small,less than 1% of flavonoids.The content of Rutin,Isoquercitrin,Hyperoside,Protocatechin and Catechins in Lizao jujube leaves are relatively highest overall.Zanhuang jujube leaves have the highest Catechin and Hyperoside,during the collection period in May Prominent is the most.The Isoquercitrin in Z.jujuba leaves reached the highest point during the September collection period.In conclusion,Z.jujuba leaves can be processed as a biologically active herbal tea.If flavonoids are used as the standard for collecting resources,the best time to pick Z.jujuba leaves is in early May.Among them,the highest content of biologically active ingredients is Lizao jujube leaves,and the second is Zanhuang jujube leaves,both of which can be used as the main raw materials for Z.jujuba leaves tea.
