In this study,based on camellia oil,the functional camellia oil products to promote brain development were created and studied by adding active substances and feedback of animal experiment results.The key technologies of microencapsulation and solubilization of water soluble or micro soluble active substances were improved.The physicochemical properties of the microcapsules were analyzed by apparatus.The functional camellia oil was prepared by adding zinc lactate microcapsules and oil-soluble active substances into refined camellia oil and its stability was studied.Through Morris water maze test,HE staining of brain tissue,Nissl staining of hippocampus,PAS staining of retina and detection of related biochemical indexes were used to explore the effect of functional camellia oil.The main results are as follows:1.Single factor and orthogonal experiment were used to investigate the impact situation and best scheme of phacoemulsification assisted preparation of zinc lactate microcapsules.The influencing elements were as follows:ultrasound time>wall material ratio>ultrasound power.The optimal combination is A2B3C3,that is,the wall material ratio is 1:1,the ultrasonic time is 20 min,and the ultrasonic power is 700 W.Under these conditions,the embedding rate was 79.12±0.43%,the drug loading was27.82±0.98%,the particle size was 161.45±2.14?m,the water content of microcapsule was 2.15±0.17%,the surface oil was 1.01±0.13%,and the total oil was33.75±0.24%.The formation of microcapsules was confirmed by electron microscopy.FT-IR showed that there was a binding effect between whey protein and maltodextrin.DSC concluded that the peak temperature of microcapsule was 102.8?,which was higher than the storage temperature of camellia oil.TG results showed that the formation of wall materials reduced the decomposition rate of microcapsules.2.The appropriate dosage of active substances was obtained by referring to various dosage standards.The optimal preparation conditions of functional camellia oil were optimized by response surface:stirring time 25 min,stirring temperature 40?,VE content 2.0 g,acid value 0.48 mg/g,peroxide value 2.12 mmol/kg.With the increase of temperature and storage time,both of av and Po V were showed a rising trend.at 10?,the av and Po V of functional camellia oil were still lower after 10 months in brown glass bottles.at higher temperature above 30?,the acid value exceeded 3 mg/g required in the standard;The shelf life of functional camellia oil was predicted to be 18months at 20?by oven method.3.The results of animal experiments showed that compared with the blank group,the low,middle and high dose groups of functional camellia oil reduced the weight of mice,and the brain index of the middle dose group had a very significant difference.Morris water maze experiments indicated that contrasted with the blank group,the escape latency of each dose group of functional camellia oil showed a downward trend,The results showed that in the functional camellia oil group,the brain tissue had complete cell structure and uniform distribution of cytoplasm,the number of cells in hippocampus increased,Nissl body was rich,retinal blood vessels were evenly distributed,and there was no distortion and aggregation phenomenon.The enzyme system in serum and liver tissue was normal.The content of malondialdehyde(MDA)in brain tissue decreased(P<0.01),while the activities of total antioxidant capacity(T-AOC)and catalase(CAT)increased(P<0.05,P<0.01),and the activity of acetylcholinesterase(a-che)decreased(P<0.05).
