Preparation Of Traditional Chinese Medicine Facial Mask Based On Rhubard And Dictamini Cortex And Study On Its Therapeutic Effect On Acne In Rabbit Ears

Traditional Chinese medicine（TCM）facial mask is an external preparation of traditional Chinese medicine with obvious bacteriostatic effect.In order to achieve the purpose of clinical treatment of acne by TCM facial mask,the preparation process optimization,quality control and anti-acne effect of TCM facial mask with rhubarb and dictamini cortex particles as the main components were studied.These studies can provide experimental basis for the development and application of new preparation of TCM facial mask.This paper mainly includes the following contents:1.In this study,rhubarb,dictamini cortex and sulfur were used as the main raw materials to make two kinds of traditional Chinese medicine facial masks（A and B）.Taking the moisture retention rate and viscosity as indexes,the preparation technology of traditional Chinese medicine facial mask was optimized by single factor experiment,and the physical and chemical indexes,stability,sensory evaluation and bacteriostasis of facial mask were studied.The results showed that the two masks were brown and yellow,with no bad smell and uniform appearance.The two masks have good heat resistance and cold resistance,the p H meets the national standard,the drying time is in the range of1080～1200s,the viscosity is close to that of the commercial masks,and the microbial indicators meet the standard.After storing for 0.5h,the moisture retention rate of the two masks is higher than 85%.The two masks were stored for 84 days,and the Total Plate Count and the Molds and Yeast Count met the standard.By measuring the bacteriostatic diameter by oxford cup,it was found that the two masks had inhibitory effects on three acne-causing bacteria.2.Established the quality standard of traditional Chinese medicine facial mask.According to the Chinese Pharmacopoeia（2020 edition）,the quality standards of active pharmaceutical ingredients（APIs）and traditional Chinese medicine masks were studied,and the contents of five total anthraquinones and fraxinone in APIs and traditional Chinese medicine masks were determined by HPLC.This time,the total content of five rhubarb anthraquinones in rhubarb granules was 17.86mg/g,and the total content of five rhubarb anthraquinones in traditional Chinese medicine masks was 1.314mg/g and1.317mg/g,so the total content of five rhubarb anthraquinones in traditional Chinese medicine masks（A and B）should not be less than 1.05 mg/g according to 80% of the average value.The measured content of dictamini cortex granules is 0.3780mg/g,and that of traditional Chinese medicine facial masks is 0.0541 mg/g and 0.0534 mg/g.Therefore,according to 80% of the average value,the content of dictamini cortex granules in traditional Chinese medicine masks（A and B）should not be less than 0.04mg/g.3.Pharmacodynamic evaluation of traditional Chinese medicine facial mask.By creating the rabbit ear acne model,the effects of Chinese medicine facial mask on the apparent indexes and histopathology of the model rabbit ears were observed,and the contents of IL-6,IL-8 and TNF-α in rabbit ear tissue were determined by ELISA.The results showed that traditional Chinese medicine masks A and B could reduce the thickness of the epidermis layer of rabbit ears and the degree of hair follicle opening expansion,decrease the contents of IL-6,IL-8 and TNF-α in rabbit ears,reduce the number of pimples in acne lesions,decrease the expansion of hair follicle funnel and reduce the accumulation of hair follicle keratins.The above results showed that masks A and B could inhibit the secretion of pro-inflammatory factors and improve the inflammatory reaction,and have therapeutic effect on rabbit ear acne mode.
