Objective:Detecte the expression levels of C/N-Myc,NAMPT and SIRT1 in skin squamous cell carcinoma and basal cell carcinoma,observe and evaluate the role and biological significance of C/ N-Myc-Nampt-SIRT1 axis in the pathogenesis of non-melanocytic skin tumors.Method:Thirty cases of SCC and fifty cases of BCC in the First Hospital of Putian from 2010 to 2019 were selected and confirmed by clinical and pathological methods.The sections were stained by immunohistochemistry and the expression mode,staining rate and staining intensity of C/N-Myc,NAMPT and SIRT1 in SCC and BCC were calculated.The data were processed by statistical software,and the correlation and significance of each index were analyzed;The expression and mode of C/N-Myc,NAMPT and SIRT1 were also evaluated by taking normal skin tissue beside tumor as control group.Result:1.In SCC,C-Myc is diffuse expressed in the tumor cell nucleus(except for the central keratinized area)with a positive rate of 80.0%.N-Myc was expressed in the nuclei of mitotic tumor cells with a positive rate of 46.7%.NAMPT was expressed in cytoplasm of tumor cells with a positive rate of 90.0%.SIRT1 was expressed in the cytoplasm of tumor cells with a positive rate of 96.7%.In BCC,C-Myc was scattered in tumor cell nucleus,and the positive rate was 36.0%.N-Myc was expressed in nuclei of mitotic tumor cells with a positive rate of 76.0%.NAMPT was expressed in the cytoplasm of tumor cells with a positive rate of 74.0%.SIRT1 was expressed in the cytoplasm of tumor cells with a positive rate of 84.0%.In normal skin,C-Myc,N-Myc,NAMPT and SIRT1 were all negative.Compared with the control group,the expressions of C/N-Myc,NAMPT and SIRT1 in SCC and BCC were statistically significant.There were significant differences in the expression of C-Myc,N-Myc,NAMPT and SIRT1 between SCC group and BCC group.2.In SCC,the positive rate of C-Myc in low differentiation was 100%,was higher than that in high differentiation group(64.7%);the positive rate of N-Myc in low differentiation was 69.2%,was higher than that in high differentiation group(29.4%).3.The positive rate of C-Myc expression in SCC group was 80.0%,was higher than that in BCC group(36.0%),while the positive rate of N-Myc expression was 46.7%,was lower than that in BCC group(76.0%).The positive rates of NAMPT and SIRT1 in SCC group were 90.0% and 96.7% respectively,which were higher than those in BCC group(74.0% and 84.0%).4.In SCC and BCC,the correlation coefficients of C-Myc and N-Myc with NAMPT,SIRT1,and NAMPT with SIRT1 were p < 0.001,r = 0.526;P < 0.001,r = 0.374;P <0.001,r = 0.681;P < 0.001,r = 0.634;P < 0.001,r = 0.613.The difference was statistically significant.C/N-Myc,NAMPT and SIRT1 were positively correlated.Conclusion:1.The expression of C/N-Myc,NAMPT and SIRT1 in SCC and BCC was positively correlated,suggesting that the C/N-Myc-NAMPT-SIRT1 axis plays an important role in the development of non-melanocytic skin tumors.In addition,each molecule of the Hh-Gli-Myc-NAMPT-SIRT1 signaling pathway promotes each other in the process of NMST occurrence and development,which may increases the invasion or metastasis ability of the disease.2.C/N-Myc may be an important factor in the invasion and metastasis of poorly differentiated SCC.3.In BCC,the expression of C-Myc is weaker than that of SCC,but the expression of N-Myc is stronger than that of SCC,which may be one of the reasons why BCC is less prone to infiltration and metastasis than SCC.4.The in-depth study on the mechanism of C/N-Myc,NAMPT and SIRT1 in Hh signaling pathway will help to further explain the pathogenesis of non-melanocytic skin tumors and provide scientific data and theoretical basis for the research and development of targeted drugs.
