| Objective: The purpose of this experiment was to investigate the interventional effects of Yiqi Yangyin Huoxue Tongluo Recipe on diabetic cardiomyopathy(DCM)model rats and its effect on transforming growth factor-beta 1,TGF-β1)/ Smad3 signaling pathway in order to provide ideas for clinical prevention and treatment of DCM.Methods: Forty healthy male SD rats were selected and randomly divided into 10 normal groups(Blank group)and 30 models after adaptive feeding for 1 week.Model rats were fed with high-sugar and high-fat diets combined with multiple low-dose(30mg /kg)streptozotion(STZ)intraperitoneal injections to establish a diabetic rat model.After modeling,they were randomly divided into model group(M group),Chinese medicine group(TCM group)and trimetazidine hydrochloride group(WM group).Tablets and other corresponding drugs were administered to the stomach for 16 weeks.Serum samples were taken at the end of the 16 th week,and related blood indexes such as TGF-β1 and hydroxyproline(Hydroxyproline,Hyp)were detected;After taking out the heart,weigh it,calculate the heart weight index(HWI),save the heart tissue,observe the pathological changes of the heart tissue by Masson staining,and detect the TGF-β1 / Smad3 signaling pathway and collagen by Western Blot Protein expression in collagenⅠ(collagen I,col-I)and collagenⅢ(collagenⅢ,col-III)was detected by real-time PCR(real-time PCR)mRNA expression of the TGF-β1 / Smad3 signaling pathway.result:1.General condition of ratsRats in group M showed more drinking,more food,more urine,weight loss,less movement,laziness,aggravated clumping,yellow hair,wet,rough,shedding,tongue-like and dark purple paw nails."Yin deficiency" syndrome of "stasis and blood stasis".Compared with the M group,each treatment group improved in different degrees.2.Comparison of FBG in each group of ratsCompared with group B,FBG in group M rats was significantly increased(P <0.05);compared with group M,FBG in group T rats was significantly decreased(P <0.05);compared with group M,there was no significant difference in group W(P> 0.05);3.Comparison of serum Hyp in each groupCompared with group B,the Hyp of rats in group M was significantly increased(P <0.05);compared with group M,the Hyp of rats in each treatment group was significantly decreased(P <0.05);compared with group W,the T group was significantly decreased(P <0.05).4.Comparison of serum TGF-β1 in each group of ratsCompared with group B,TGF-β1 in M group rats was significantly increased(P <0.05);compared with M group,TGF-β1 in each treatment group was significantly decreased(P <0.05);compared with W group,T group was significantly decreased(P <0.05).5.Comparison of HWI in each experimental groupCompared with the M group,there was no significant difference in HWI between the treatment groups(P> 0.05).6.Cardiac morphology observationThe heart structure of group B was normal.Group M showed myocardial fibers arranged disorderly,myocardial interstitial collagen deposition,and collagen fiber area significantly increased.Each treatment group improved to varying degrees.7.Real-time PCR detection of TGF-β1 and Smad3 mRNA expression in myocardial tissueCompared with group B,TGF-β1 and Smad3 mRNA expression in M group rats were significantly increased(P <0.05).Compared with the M group,the expression of each treatment group was significantly decreased(P <0.05 or P <0.01);compared with the W group,the T group was significantly decreased(P <0.05).8.Western Blot detection of TGF-β1,Smad3,Col-Ⅰ,Col-Ⅲ protein expression in myocardial tissue of each group of ratsCompared with group B,the expression of rats in group M was significantly increased(P <0.05);compared with group M,the protein expression of each treatment group was significantly reduced(P <0.05);compared with group W,the group T was significantly decreased(P <0.05).Conclusion:1.Yiqi Yangyin Huoxue Tongluo Recipe can improve the general condition of DCM rats,to a certain extent,reduce blood glucose levels,relieve cardiac pathological damage,reduce myocardial fibrosis,and have obvious protective effects on DCM rat heart tissues.2.Yiqi Yangyin Huoxue Tongluo Recipe can inhibit cardiac myocardial fibrosis in DCM rats,and down-regulate the expression of TGF-β1 and Smad3 proteins,suggesting that inhibiting overexpression of TGF-β1 / Smad3 signaling pathway may protect the heart of DCM rats Mechanism of action.3.In the prevention and treatment of diabetic myocardial fibrosis,Yiqi Yangyin Huoxue Tongluo Recipe and trimetazidine hydrochloride both have curative effects,but from the current experimental data analysis,the effect of traditional Chinese medicine prescription is better than trimetazidine tablets. |
