Objective: Use herbal cake-separated moxibustion to intervene in atherosclerotic rabbits,observe the effect of herbal cake-separated moxibustion on atherosclerotic rabbit liver TGR5 and serum inflammation-related factors TNF-?,IL-6,IL-1?,to explore the mechanism of herbal cake-separated moxibustion to prevent and treat atherosclerosis,and to provide scientific basis for the prevention and treatment of atherosclerosis with herbal cake-separated moxibustion.Methods: 32 New Zealand male rabbits were randomly divided into blank group,model group,herbal cake-separated moxibustion and atorvastatin group,with 8rabbits in each group.Atherosclerotic rabbit model was established by feeding with high cholesterol and propylthiouracil(10mg/kg·d),Two groups of acupoints,one was“Juque”(CV 14),double“Tianshu”(ST 25)and double“Fenglong”(ST 40),the other one was double“Xinshu”(BL 15),double“Ganshu”(BL 18)and double“Pishu”(BL 20),The duration of continuous moxibustion at each point is about30 minutes,The two groups were treated with moxibustion alternately,once a day for twelve successive weeks.Observation of the structure of the rabbit thoracic aorta under a 400 x optical microscope,detection of blood lipid levels in each group of rabbits,measure the expression of liver TGR5 by Quantitative Real-time PCR,Elisa detects the expression of TNF-?,IL-6 and IL-1? in rabbit serum.Results:1.HE staining: in the blank group,the internal structure of abdominal aorta was intact,the intima was smooth,complete,continuous,uniform in thickness,without lipid infiltration and obvious lipid spots,the intima of the abdominal aorta in the model group was obviously thickened,unevenly protruding into the lumen and a large number of foam cells were formed.The intima of the herbal cake-separated moxibustion group and the atorvastatin group were still clearly visible,and the endothelium of the tube wall had continuity,The foam cells are less than the model group.2.Blood lipid level: compared with the blank group,the rabbit serum TC,TG,HDL-C and LDL-C levels in the model group increased(P<0.01).Compared with the model group,the serum levels of TC,TG,HDL-C,and LDL-C in the herbal cake-separated moxibustion group and the atorvastatin group decreased(P<0.01 or P<0.05).3.TGR5 of liver: compared with the blank group,the expression of TGR5 m RNA in the model group decreased(P< 0.05).Compared with the model group,the expression of TGR5 m RNA in the herbal cake-separated group and the atorvastatin group increased significantly(P<0.05).4.Serum inflammation-related factors: compared with the blank group,the expression of TNF-?,IL-6 and IL-1? in the model group increased(P<0.05),compared with the model group,the expression of TNF-?,IL-6 and IL-1? in the atorvastatin group decreased(P<0.05),and the expression of IL-1? in the herbal cake-separated moxibustion group decreased(P<0.05).The expression of TNF-? and IL-6 in herbal cake-separated moxibustion group decreased(P>0.05).Conclusion:1.Herbal cake-separated moxibustion can effectively regulate blood lipid levels,improve the damage of the arterial wall,prevent the production of foam cells and inhibit the formation of AS plaque.2.Herbal cake-separated moxibustion can activate TGR5,inhibit the expression of serum TNF-?,IL-6,IL-1?,reduce inflammatory cell infiltration,inhibit the formation of sclerotic plaque and inflammation within the plaque,thereby reducing AS disease.
