Schisandra sphenanthera Rehd.Et Wils.,belonging to Schisandra genus Schisandraceae,has been listed as Grade ? protected wild medicinal materials in China.Triterpenoids are one of the main pharmacological components of S.sphenanthera,which have antioxidant and bacteriostatic effects.S.sphenanthera is a woody lianas.During the wild cultivation,a large number of stems and leaves are discarded after pruning branches,and the stems and leaves contain a variety of active substances.Therefore,this experiment uses different periods of S.sphenanthera(germination,leaf development,flowering,and deciduous)Stage)stems and leaves are used as materials.Response surface methodology optimizes the ultrasonic-assisted extraction of total triterpenoids of S.sphenanthera,and compares the content of total triterpenes in different parts of different periods;Separation and purification of total triterpenes of S.sphenanthera by D-101 type macroporous adsorption resin;through antioxidant and reducing power testing,compare the anti-oxidation ability of the total triterpenoids of S.sphenanthera in different periods at different periods;use 4 kinds of bacteria and 1 kind of fungus to test the antibacterial ability of S.sphenanthera.The main conclusions are as follows:1.Optimization of ultrasonic-assisted extraction process for stems and leaves of S.sphenanthera by response surface methodology.The optimal extraction conditions of stems were as follows:material-to-liquid ratio of 1:30 g/mL,methanol concentration of 78%,ultrasonic time of 52 min,and the extraction yield was 1.14±0.23%.The optimal extraction conditions of leaves were as follows:material-to-liquid ratio of 1:21 g/mL,methanol concentration of 91%,ultrasonic time of 76 min.The extraction yield was 1.78±0.17%,and the theoretical value was basically consistent with the actual value.2.The content of total triterpenoids in the stems of S.sphenanthera was 7.47-15.18 mg/g,the highest at germination stage and the lowest at deciduous stage.The content of total triterpenoids in leaves was 15.47-19.66 mg/mL,the highest at flowering stage and the lowest at deciduous stage.The content of total triterpenoids in leaves was higher than that in stems.3.D-101 type macroporous resin purifies the total triterpenes of S.sphenanthera.The purity of the total triterpenes of the stem and leaves before purification is 26.33±1.18%,26.30±13.09%,and the purity of the total triterpenes after purification is 75.04±3.63%,75.00±4.02%,the purity is increased by about 2.8 times.4.The total triterpenoids of S.sphenanthera have a strong overall antioxidant capacity,and there is an obvious dose-effect relationship.The total triterpenoids of stem and leaves have the scavenging ability of free radicals at flowering stage>leaf development stage>budding stage>deciduous stage,except for the scavenging ability of stem total triterpenes on ·O2-free radicals,the scavenging capacity of stem and leaf total triterpenes is leaf development stage>flowering stage>budding stage>fallen leaves period.The reducing power of total triterpenoids in the stems and leaves of S.sphenanthera is stronger than that of VE and weaker than that of VC.The antioxidant capacity of the stems of S.sphenanthera is stronger than that of the leaves,except for the deciduous period(removal rate of DPPH and ·OH),the germination period(DPPH),and the flowering period(·O2-).5.In vitro antibacterial experiments showed that the total triterpenoids from the stems and leaves of S.sphenanthera had stronger antibacterial activity against Bacillus subtilis and Pseudomonas aeruginosa than against Staphylococcus aureus and Escherichia coli,but had no antibacterial activity against Candida albicans(fungi).The minimum inhibitory concentration(MIC)was 0.63-5.00 mg/mL,the minimum bactericidal concentration(MBC)was 2.50-20.00 mg/mL,and the growth rate and cycle of bacteria were effectively inhibited.6 After purification,there is no correlation between the total triterpene content in the stem and leaf of S.sphenanthera in different periods and its antioxidant and antibacterial capacity.There is a negative correlation between antioxidant and antibacterial capacity,among which the ability to scavenge DPPH free radicals and the ability to inhibit B.subtilis,there was a very significant negative correlation(p<0.01),and the ability to inhibit E.coli and P.aeruginosa was significantly negatively correlated(p<0.05);the ability to scavenge ·OH free radicals was correlated with the inhibition of B.subtilis and P.aeruginosa.significantly negative correlation(p<0.05).In addition,this experiment found that the ability to scavenge DPPH free radicals was extremely positively correlated with the ability to scavenge ·OH free radicals(p<0.01).7.Based on the comparison of the antioxidant and bacteriostatic abilities of other active components,the stems and leaves of S.sphenanthera at leaf spreading and flowering stages are more suitable for the treatment of oxidative damage induced by·O2-and·OH radicals and related diseases caused by S.aureus and other bacteria.In summary,through the optimization of the extraction process of the total triterpenoids of S.sphenanthera,the content determination of different parts in different periods,purification and separation,and the study of antioxidant and antibacterial activities.The research lays the foundation for further research on the total triterpenoids of S.sphenanthera and its biological activity,and provides data support for alleviating the tension of fruit resources on the side and strengthening the effective use of other parts.
