| With the improvement of living standards,digestive system diseases are gradually increasing,and the health of intestinal microecology is particularly important.In order to improve the effect of Hawthorn and malt on promoting digestion and probiotics to maintain the balance of intestinal flora,Hawthorn,malt and probiotics were used as research objects to extract effective substances from Hawthorn and malt by water extraction.the probiotics were expanded and cultured,and the relieving effect of Hawthorn malt water extract and probiotics on food accumulation in mice with high protein diet was studied by animal experiment.At the same time,the water extracts and probiotics were freeze-dried by ultra-low temperature freeze-drying to obtain the probiotic hawthorn malt solid beverage,and the co-intervention of hawthorn malt solid beverage and probiotics was carried out to alleviate the food accumulation in mice with high protein diet through animal experiments.effect.The main research contents and conclusions are as follows:(1)Water extract of Hawthorn malt and its recombination with probiotics to alleviate food accumulation in mice fed with high protein dietThe hawthorn and malt are extracted by water extraction to obtain the hawthorn malt water extract;Lactobacillus casei LH23(Lactobacillus casei LH23)is selected as the probiotic,and the probiotic bacteria suspension is obtained by separation,purification and further activation and cultivation.The hawthorn malt water extract and its complex with probiotics have been verified by animal experiments.Compared with the model group,it can increase the number of defecation grains and the water content in the feces of the mice with phagocytosis caused by high-protein diet(p<0.01).The gastric residual rate decreased to varying degrees(p<0.05),and the small intestinal propulsion rate increased significantly(p<0.01);increased the secretion of gastrin(GAS)in gastric antrum tissue(p<0.05),and decreased the growth rate The secretion of somatostatin(SS)(p<0.05);the inflammatory factors interleukin-10(Interleukin-10,IL-10)and tumor necrosis factor-α(tumornecrosis factor-α,TNF-α)were different in small intestinal tissue degree of rise and fall.In addition,the effects of hawthorn malt water extract and its complex with probiotics on the content of short-chain fatty acids in the colon content of mice with hyperphagia induced by high protein diet were analyzed by gas chromatography.The results showed that Lactobacillus casei LH23 group,hawthorn malt water extract After dietary intervention in the group,hawthorn malt water extract and probiotics complex group,the content of acetic acid(p<0.05)in the intestinal tract of mice was increased,and the production capacity of isobutyric acid was decreased,but due to the differences between individual mice,without a significant difference.(2)Research and optimization of conditions of hawthorn malt water extract and probiotic complex solid beverage40min was extracted from hawthorn&malt in 2L water at 1:5 to get water extract of900ml,then 60min was concentrated at 60℃by rotary evaporator to get 150ml concentrate,and then vacuum freeze-dried plate was placed for 12h to get about 1.2g freeze-dried powder.The ratio of material to liquid(g/ml)was 166.67g freeze-dried powder equivalent to 10g raw material,including 1.667g Hawthorn and 8.33g malt.The number of live Lactobacillus casei LH23 was 1.54*10~9CFU/ml.Single factor experiments showed that4%trehalose was the best freeze-drying protectant,the number of live bacteria after freeze-drying was 1.423*10~9CFU/ml,and the freeze-drying protection rate was 92.4%.Water extract freeze-dried powder and freeze-dried bacterial powder were sifted through 60mesh and then mixed according to 1.5g:1g ratio to obtain solid beverage.(3)Co-intervention of hawthorn malt solid drink and probiotics on food accumulation in mice with high protein diet miceBoth hawthorn malt freeze-dried extract and compound solid beverage can significantly increase the water content of feces in mice with phagocytosis caused by high-protein diet(p<0.001),reduce the residual rate in the stomach of mice with phagocytosis(p<0.01),and significantly increase the rate of intestinal propulsion(p<0.01).At the same time,the secretion of gastrin(GAS)in gastric antrum tissue was increased(p<0.01),and the secretion of somatostatin(SS)was decreased overall(p<0.05),due to differences between individual mice,there is no significant difference.In addition,no tissue damage was found in gastric antrum and small intestine tissue sections. |
