Arabinogalactan extracted from Larix wood has rich biological activity.It was used as a food additive in the past.Now its pharmacological activity is gradually known.Arabinogalactan has been applied to the pharmaceutical industry,but there is little research on functional products of arabinogalactan at home and abroad.In view of this,the extraction of arabinogalactan from Larix olgensis Henry sawdust was studied by using a cellulase-assisted citric acid-sodium citrate buffer system.The extraction process was optimized by single factor and response surface methodology.In order to further develop and utilize arabinogalactan,arabinogalactan buccal tablets and arabinogalactan solid beverages were prepared by adding suitable pharmaceutical excipients into the extracted arabinogalactan.The quality control and stability of arabinogalactan buccal tablets were investigated.The main conclusions are as follows:(1)By single factor and response surface methodology,the extraction technology of arabinogalactan from larch was optimized.Factors affecting the extraction rate are as follows: Enzyme addition(A)> Temperature of extraction(C)> Time of extraction(B)> Material-liquid ratio(D).And the best extraction process is as follows: the additional amount of enzyme was 1%,the time of extraction was 67 min,the temperature of extraction was 58℃,Material-liquid ratio was 1:18 g/m L.Under the optimum technological conditions,three verification experiments were carried out.The extraction rate of arabinogalactan was 21.95±0.52%,with little difference from the predicted results.The results showed that the optimized extraction process was reasonable and feasible.(2)Arabinogalactan was added with some pharmaceutical excipients to prepare the buccal tablets.Firstly,the types and proportions of arabinogalactan,filler,disintegrant,corrective,and lubricant were determined through single factor experiment,and through an orthogonal experiment,the best preparation technology of arabinogalactan buccal tablets was determined.The additional amount of arabinogalactan is 25.0%,the additional amount of microcrystalline cellulose and mannitol(1:3)is 66.6%,the additional amount of citric acid as a flavoring agent is 2.0%,the additional amount of croscarmellose sodium as a disintegrant is 5.0%,the addition amount of antioxidant is 0.6%,and the addition amount of magnesium stearate as a lubricant is 0.8%.The appearance,disintegration time,hardness,friability,and quality difference of arabinogalactan buccal tablets prepared by the optimal process were tested,and the results met the relevant standards,which proved that the process was reasonable and feasible.(3)The influencing factors and stability of arabinogalactan buccal tablets were studied.The results showed that all the indexes of arabinogalactan buccal tablets met the requirements during the test period,and the arabinogalactan buccal tablets were sealed and packaged in white bottles with moisture-proof particles.In vitro,antioxidant experiments showed that arabinogalactan and arabinogalactan buccal tablets had good scavenging capacity on DPPH free radicals and ABTS free radicals,while arabinogalactan buccal tablets had better hydroxyl radical scavenging capacity and iron ion reducing capacity than arabinogalactan,which may be due to the raw materials of buccal tablets enhanced the antioxidant capacity of buccal tablets.Therefore,it is a feasible way to prepare arabinogalactan tablets with appropriate pharmaceutical excipients.(4)The sensory evaluation of arabinogalactan solid beverage was carried out by the fuzzy mathematics evaluation method.A single-factor experiment was conducted to study the effects of the amount of arabinogalactan,mannitol,mint powder,inulin,and malic acid on the sensory evaluation of solid drinks.The optimal formula was as follows: arabinogalactan 17.5%,mannitol 28.2%,peppermint powder 18.9%,inulin34.1%,and malic acid 1.1%.Based on the optimum technology,the moisture,molding rate,solubility,fluidity,and total sugar content of the arabinogalactan solid beverage were determined.The results were in line with the relevant standards of solid beverages in China.
