| 80 Yanbian yellow cattle were used as experimental materials.The SNP of LPL gene and WFIKKN2 gene were detected by PCR and HRM,respectively.The genetic effect of the experimental population was analyzed,and the correlation between LPL gene and WFIKKN2 gene was analyzed.Finally.The possibility of LPL and WFIKKN2 gene as candidate gene for beef quality traits of Yanbian yellow cattle were discussed to improve the beef quality of Yanbian yellow cattle,as well as to provide theoretical basis for the selection and improvement of Yanbian yellow cattle.The test results are as follows:1.Five polymorphic loci of LPL gene in Yanbian yellow cattle were analyzed by high resolution fusion curve and sequenced subsequently.The g.6215A>G loci of LPL gene showed two genotypes:AA and AG,and the dominant allele was A;There were two genotypes at g.18341C>T locus,CC and CT,C is the dominant allele;HRM at 18570bp locus shows no genotyping;there were two genotypes at g.355427T>A,AA and AT,A was the dominant allele;c.322G>A,HRM shows no genotyping.All of the five polymorphic loci showed moderate polymorphism and were in Hardy Weinberg genetic balance.2.HRM analysis showed that there were three polymorphic loci in genotyping,which were correlated with meat quality traits of Yanbian yellow cattle.g.6215A>G loci were significantly correlated with carcass weight,fat content,protein content and water content of Yanbian yellow cattle,which were:AG carcass weight and fat content were significantly higher than AA;AA protein content and water content were significantly higher than AG;g.18341C>T locus and live weight before slaughter,carcass weight and backfat thickness of Yanbian yellow cattle,which showed that:CT type was significantly higher than CC type;g.355427T>A locus was higher than AA type in live weight before slaughter,backfat thickness and fat content.3.By PCR direct sequencing,there was a SNP locus with moderate polymorphism in WFIKKN2 gene,which was in Hardy Weinberg genetic balance.The results showed that the g.1170A>C locus of WFIKKN2 gene was significantly correlated with carcass weight,fat content,water content and backfat thickness of the tested yellow cattle,and showed that AA type was significantly higher than AC type,and had no significant difference with protein content and live weight before slaughter. |
